辛伐他汀抗血脂代谢紊乱型大鼠血管平滑肌细胞增殖的实验观察

摘    要:目的 观察辛伐他汀对血脂代谢紊乱型大鼠血脂水平的影响以及对血管平滑肌细胞增殖的抑制作用。方法 将 4 0只Wister大鼠分为空白对照组 ,模型组 ,辛伐他汀组 3组 ,喂饲高脂饲料造模 ,辛伐他汀组每天用西药辛伐他汀 (1.0ml 10 0g体重 ,1ml含药 0 .17mg) (浙江新昌京新制药有限公司 ,批号 :0 0 0 1175 )灌胃 ,以上各组共灌胃 10周 ,自由饮水 ,于实验第 77天全部处死取材。分别检测各组大鼠的总胆固醇 (TC) ,甘油三酯 (TG) ,低密度脂蛋白胆固醇 (LDL C) ,高密度脂蛋白胆固醇 (HDL C)的含量 ,用PCNA标记法检测VSMC增殖率。结果 模型组的TC水平明显高于其他两组 (P <0 .0 1) ,LDL C的含量空白对照组和模型组差异有显著性。模型组的PC NA阳性细胞率明显高于其他两组 (P <0 .0 1)。经相关性分析显示 ,模型组VSMC增殖率与血清TG、TC含量呈正相关 ,其相关系数分别为r=0 .786 ,r=0 .6 96 ,经检验差异有显著性 (P <0 .0 1)。结论 西药辛伐他汀抑制内源性胆固醇的合成 ,为血脂调节剂 ,它通过降低血脂异常大鼠的TC含量 ,降低LDL C水平 ,还可能通过防止脂质的氧化 ,改善内皮功能 ,恢复内皮依赖性血管舒张反应 ,抑制VSMC的增生与迁移

Abstract:Objective To observe the relation between blood lipids abd proliferation of vascular smooth muscle cells (VSMC) and find the possible mechanism and precaution of atherosclerosis.Methods Fourty male rats were divided randomly into three groups,group control,model lipasicmixture and simvastation.The group lipasic mixture,1.1ml/100g lipsic mixture was added to forage with cholesteral and lipid,to the group station,0.1ml/100g Simvastation was added to forage with cholesterol and lipid,to group control,ordinary forag only.Biochemical parameters and the parcent of proliferating cell nuclear antigen (PCNA) of VSMC were carried out 10 weeks later.Results Forage with cholesterol and lipidincreased the level of TC,TG and LDL-C in group model.The positive ratio of PCNA of grroup model is higher than the others (P<0.01).Conclusion Simvastation can inhibit experimental hyoerlipemia and atherogenesis.It is an ideal and effective medicine in preventing and treating hyperlipemia and atherogenesis.
