大鼠急性心肌梗死后心肌组织中miR-214的表达变化

摘    要:目的观察大鼠心肌梗死后心室重塑过程中心肌组织中miR-214的表达变化。方法 10只雄性SD大鼠,通过前降支动脉结扎法建立大鼠急性心肌梗死模型,模型制备成功后随机分为心肌梗死后14 d组、28 d组,另设对照组,每组5只动物。抽提梗死周边区心肌组织的RNA及miRNA,应用荧光定量PCR法检测MHC-α、MHC-βmRNA及miR-214的表达。结果心肌组织中MHC-αmRNA表达随着心肌梗死时间延长而逐渐降低。与假手术组比较,心肌梗死术后14 d组MHC-α的表达下降46%(0.54±0.06 vs.1.00±0.09,P<0.01),心肌梗死术后28 d组MHC-α的表达下降63%(P<0.01)。心肌组织中MHC-βmRNA表达随着心肌梗死时间延长而逐渐增加。与假手术组比较,心肌梗死术后14 d组MHC-β的表达增加2倍(P<0.01),心肌梗死术后28 d组MHC-α的表达增加3.22倍(P<0.01)。心肌梗死术后14 d组和28 d组梗死周边区心肌组织中miR-214的表达逐渐增加。与假手术组相比,心肌梗死术后14 d组miR-214的表达增加33%(P<0.01),心肌梗死术后28 d组miR-214的表达增加88%(P<0.01)。结论大鼠心肌梗死后心室重塑过程中心肌组织中miR-214的表达上调,miR-214可能参与调控了心肌梗死后心室重塑。

Abstract:Objective To detect the change of expression level of miR-214 in myocardium post acute myocardium infarction in rat.Method: Rats underwent left descending coronary ligation or sham surgery.Rats with MI was assigned in two groups(n=5).Real-time PCR was used to detect the expression of miR-214,MHC-α,and MHC-βmRNA in myocardium.Results The expression level of miR-214 in myocardium was up-regulated in the 14th and 28th day post MI.Compared to sham group,the expression level of miR-214 in myocardium increased by 33%(P〈0.01) in the 14th day and by 88%(P〈0.01) in the 28th day post MI.The expression level of MHC-α,MHC-β mRNA altered in the progress of ventricular remodeling post MI in rat.the expression level of MHC-α mRNA in myocardium decreased by 46%(P〈0.01) in the 14th day and by 63%(P〈0.01) in the 28th day post MI.The expression level of MHC-βmRNA in myocardium increased by 2 fold(P〈0.01) in the 14th day and by 3.22 fold(P〈0.01) in the 28th day post MI.Conclusion The expression level of miR-214 altered in the progress of ventricular remodeling post MI in rat.MiR-214 might be involved in ventricular remodeling post MI.
