依达拉奉对L5脊神经结扎模型大鼠神经组织氧化损伤的保护作用研究

摘    要:目的:研究依达拉奉对L5脊神经结扎模型大鼠神经组织氧化损伤的保护作用。方法:取大鼠随机分为假手术+药物(依达拉奉3mg·kg-1)组、手术对照组、手术+药物(依达拉奉3mg·kg-1)组,每组18只。第1组行假手术,后2组行L5脊神经结扎,术后立刻腹腔注射相应药物,每天2次(时间为10:00、16:00),连续给药3d。考察术后第3、6、12、24、48、72h各组大鼠脑组织中超氧化物歧化酶(SOD)、一氧化氮(NO)、丙二醛(MDA)含量变化。结果:与假手术+药物组比较,手术对照组和手术+药物组各时间点大鼠脑组织中SOD含量均明显减少;除手术+药物组48、72h外,其他各时间点NO、MDA含量均明显增加(P<0·05或P<0·01)。与手术对照组比较,手术+药物组各时间点大鼠脑组织中SOD含量均明显增加,NO、MDA含量均明显减少(P<0·05或P<0·01)。结论:依达拉奉对L5脊神经结扎模型大鼠神经组织的氧化损伤具有一定的保护作用,可能与其能有效清除氧自由基有关。

Abstract:OBJECTIVE:To investigate the protective effects of edaravone on oxidative damage of neurological tissue in rats after L5 spinal nerve ligation.METHODS:Rats were randomly divided into sham operation plus drug group(edaravone 3 mg·kg-1),operation control group and operation plus drug group(edaravone 3 mg·kg-1) with 18 rats in each group.The first group received sham operation,and the latter two groups received L5 spinal nerve ligation.They were given intraperitoneal injection of relevant drugs immediately after operation twice a day(10:00,16:00) for consecutive 3 days.The contents of SOD,NO and MDA in cerebral tissue were detected in 3 h,6 h,12 h,24 h,48 h,72 h after operation.RESULTS:Compared with sham operation plus drug group,the contents of SOD in cerebral tissue in operation control group and operation plus drug group were decreased significantly;the contents of NO and MDA in operation plus drug group increased significantly at above time points except for 48 h and 72 h(P<0.05 or P<0.01).Compared with operation control group,the contents of SOD in operation plus drug group increased significantly at above time points while the contents of NO and MDA decreased significantly(P<0.05 or P<0.01).CONCLUSION:Edaravone can effectively protect neurological tissue against oxidative damage in rats after L5 spinal nerve ligation,which is associated with removing oxygen free radicals.
