白介素-6对马法兰诱导多发性骨髓瘤细胞KM3凋亡的影响

摘    要:目的探讨白介素-6(IL-6)在马法兰诱导人多发性骨髓瘤细胞株KM。凋亡效应中的作用及其可能的分子机制。方法运用流式细胞仪技术(FACS)、DNA片段化百分率、凋亡细胞的原位检测(TUNEL法)和Western blot分析,观察II;6对马法兰诱导KM。细胞凋亡的影响及其caspase-3、caspase-8蛋白表达的变化。结果IL-6作用可使马法兰诱导的KM3细胞凋亡减少,同时caspase-3蛋白表达降低。结论1176可通过调节caspase-3、caspase-8表达保护马法兰诱导的KM3细胞凋亡。

Abstract:Objective To explore the robe of interleukin-6 in the apoptosis of multiple myeloma (MM) cell lines KM_3 induced by melphalan and its molecular mechanism. Methods Apoptosis was confirmed by flow cytometry, DNA fragmentation rate and TUNEL. The expression of caspase-3, caspase-8 proteins in KM_3 cell, which was assessed by Western blot analysis, after melphalan treatment and cultured with or without interleukin-6. Results KM3 cells in the presence of interleukin-6 showed lower rate of apoptosis compared with that in the absence of interleukin-6 (P<0.05). Also levels of caspase-3, caspase-8 expression were lower accordingly. Conclusion Interleukin-6 affects the sensitivity of the KM3 cell to melphalan induced apoptosis, and it can prevent cells from apoptosis by down-regulating the expression of caspase-3.
