昆明山海棠碱诱导Jurkat淋巴瘤细胞膜磷脂酰丝氨酸外翻及瘤细胞超微结构改变的研究

摘    要:目的 检测昆明山海棠碱(THH碱)诱导Jurkat T淋巴瘤细胞的凋亡效应。方法 THH碱多剂量多时间点诱导Jurkat T淋巴瘤细胞细胞后,采用Annexin-V-Fluorescence标记膜外翻磷脂酰丝氨酸(Phosphatidylserine,PS)的凋亡细胞,同时荧光素PI标记坏死细胞,流式细胞术检测分别检测其阳性细胞率。同时采用判断凋亡发生的“金标准”——形态学观察方法观察其超微结构的改变。结果 THH碱诱导后磷脂酰丝氨酸外翻的细胞显著增加,有明显的时间和剂量相关性,超微结构观察可见大量细胞明显发生细胞体积缩小、核固缩、胞浆浓缩、凋亡小体形成等凋亡典型的形态特征。结论 THH碱能非常显著地诱导Jurkat T淋巴瘤细胞发生凋亡。

Abstract:Objective To detect the apoptotic effect of Tripterygium Hypoglaucum(Levl) Hutch (THH) alkaloids on Jurkat T lymphoma cells. Methods After Jurkat cells were induced by THH alkaloids at different time points and different concentrations, translocated phosphatidylserine(PS) was labeled with Annexin V Fluorescence and the dead cells labeled with propidium iodide(PI). PS translocated cells and dead cells were measured with flow cytometry. The morphological changes, including ultrastructural changes, were observed. Results THH alkaloids could effectively induce the translocation of phosphatidylserine in time and dose dependent manners. Observation of the ultrastructural changes showed that there were a series of typical apoptotic changes, including decreased cell volume, apoptotic bodies, pyknosis and condensed cytoplasm. Conclusion THH alkaloids can effectively induce the apoptosis of Jurkat T lymphoma cells.
