糖脂平对胰岛素抵抗大鼠骨骼肌氧化应激的影响

摘    要:目的建立喂养型胰岛素抵抗(insulinresistance,IR)大鼠模型,观察糖脂平对喂养型IR大鼠氧化应激的影响。方法选用体重在180~200g的8周龄雄性SD大鼠60只,适应性喂养一周后,随机分为空白对照组15只和造模组45只。空白对照组采用普通饲料喂养,造模组采用高脂饲料喂养。8周造模成功后,将造模组随机分为:模型组、糖脂平组、罗格列酮组各15只。糖脂平组给予糖脂平按生药20g/kg·d灌胃;罗格列酮组给予罗格列酮0.8mg/kg·d灌胃;模型组和空白对照组灌服等量的生理盐水;末次给药后禁食12小时,用正常血糖胰岛素钳夹技术评价胰岛素敏感性,检测各组大鼠的体重、血糖、血清胰岛素水平,并留取骨骼肌,检测组织匀浆中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH—Px)、丙二醛(MDA)、过氧化氢酶(CAT)等指标。结果糖脂平能增加胰岛素敏感性,改善胰岛素抵抗,糖脂平组的M值显著高于模型组,体重、血糖、胰岛素均低于模型组,差异有统计学意义(P〈0.05),与罗格列酮组比较,体重一项有统计学意义(P〈0.05),血糖与胰岛素无统计学意义(P〉0.05);糖脂平组SOD、GSH—Px、CAT水平显著高于模型组,MDA显著低于模型组,差异有统计学意义(P〈0.05),与罗格列酮组比较,差异无统计学意义(P〉0.05)。结论糖脂平可改善胰岛素抵抗,也可改善氧化应激,反应了糖脂平可以通过改善胰岛素抵抗的上游通路——氧化应激来提高机体代谢活力,改善胰岛素抵抗,其作用机制可能是通过改善骨骼肌中线粒体的活性氧。

Effects of Tangzhiping on oxidative stress in rat insulin resistance model

Abstract:Objective To investigate the effects of Tangzhiping on oxidative stress in rats with in- sulin resistance. Methods 60 male SD rats (weighs 180 -200 g, ages 8 w) were selected and fed a week for adaptation. Then, the rats were randomly divided into blank control group ( n = 15 ) and model group ( n = 45 ). Blank control group were fed with common food, while model group were fed with high fat food, last for 8 weeks. After 8 weeks the mould was successfully made, these modules were randomly divided in- to : the model group, Tangzhiping group, rosiglitazone group ( n = 15 in each group). Tangzhiping group were given Tangzhiping formula particles according to the statement 20 g/kg · d to fill in the stomach; Ros- iglitazone group were given 0. 8 mg/kg · d rosiglitazone gavage; Model group and blank control group were given the same amount of saline solution, again for 8 weeks ; 12hs after the last administration of fasting, levels of insulin sensitivity were evaluated by the "euglycemic hyperinsulin clamp technique", and levels of body weight, fast plasma glucose, serum insulin , were observed, Meanwhile superoxide dismutase ( SOD), glutathione peroxidase (GSH-Px) malonaldehyde ( MDA ), and catalase (CAT) of skeletal muscle were measured. Results The M level was significantly higher while mean body weight, blood glucose, and fasting insulin of Tangzhiping group were lower than that of model group,. The differences were statisti- cally significant ( P 〈 0. 05 ), with rosiglitazone group, which body weight was statistically significant ( P 〈 0.05) , blood sugar and insulin had no statistical significance (P 〉 0. 05) ; The mean SOD, GSH and CAT level of Tangzhiping group is significantly higher than model group, MDA level was lower, the differences were statistically significant ( P 〈 0. 05 ), with rosiglitazone group statistically significant ( P 〈 0. 05 ). Con- clusion Tangzhiping can improve insulin resistance, and also improve oxidative stress, which reflects that Tangzhiping can enhance body's metabolic activity through improving the upstream pathway of IR--oxidative stress, in the end improve IR. Its mechanism may be through improving skeletal muscle mitochondrial ROS.
