摘    要: 目的:建立血中华法林浓度的毛细管胶束电动色谱分析方法,并对其进行评价。方法:用二氯甲烷提取血清中华法林,5-乙基-5-P-甲苯基巴比妥酸为内标,操作电压15 kV,210 nm波长检测。电泳缓冲液由25 mmol/L硼酸盐,25 mmol/L十二烷基硫酸钠,10 mmol/L氯化钠(pH 9.0)组成。结果:日内RSD1.4%~6.2%,日间RSD1.4%~4.7%,在0~10 μg/ml范围内呈线性,最低血清华法林检测浓度为0.03 μg/ml。测定结果与反相高效液相色谱法进行了比较,两种方法有很好相关性,r=0.996 7(P>0.1)。结论:毛细管胶束电动色谱法精密度、准确度高,专一性强,是一种较理想的血药浓度检测方法。

Abstract:OBJECTIVE:To evaluate a selective procedure for the determination of warfarin in serum by micellar electrokinetic capillary chromatography.METHODS:Extraction of the drug with dichlormethane and 5-ethyl-5-p-tolylbarbituric acid was used as internal standard,applied voltage 15 kV,detected at 210 nm wavelength.The run buffer contained 25 mmol/L borate,25 mmol/L sodium dodecylsulphate,and 10 mmol/L sodium chloride(pH9.0).RESULTS:The precision of the method,expressed as relative standard deviation,with in-day ranged from 1.4% to 6.2%,and day to day from 1.4% to 4.7%.The method was linear from 0 to 10 μg/ml and the limit of detection was 0.03μg/ml.Compared with those obtained by HPLC,the concentration of warfarin obtained by the two methods had good linear relationship with a slope of 0.997 5 and a correlation cofficient of 0.996 9(P>0.1).CONCLUSION:The method showed a good precision and accuracy,and is suitable for determination of warfarin in serum.
