枸杞多糖对大鼠骨折愈合过程中BMP-2表达的影响

摘    要:目的研究枸杞多糖(LBP)对SD大鼠股骨骨折愈合过程中骨形态发生蛋白-2(BMP-2)表达的影响,探讨枸杞多糖应用于骨折治疗的可能性。方法选用3月龄雄性SD大鼠90只,随机分为三组:LBP高剂量组(H组,160mg.kg-1.d-1)、LBP低剂量组(L组,40mg.kg-1.d-1)、蒸馏水组(W),每组30只。制备右侧股骨干骨折模型,行克氏针髓腔内固定。造模术后分别给药物干预,于术后第2、4、6周每组各取10只大鼠通过免疫组织化学技术检测骨痂组织中BMP-2表达情况,并做相应HE染色,对照观察骨痂组织细胞演变及BMP-2表达变化。结果免疫组化显示各组BMP-2表达在骨折后第2、4、6周均持续升高,各时间点H组表达强度均高于L组和W组,各时间点H组BMP-2表达的MOD值均高于L、W组(P<0.05);与HE染色切片对照观察,各组表达BMP-2阳性细胞种类相同,随骨折愈合进程的增加,BMP-2在成纤维细胞、软骨细胞和骨小梁内的骨细胞表达逐渐减少,在成骨细胞一直呈强阳性表达。组织切片显示H组骨折愈合的组织学演变要早于L组和W组。结论 LBP能够促进骨折愈合过程中BMP-2的表达,存在量-效关系,对骨折愈合有一定的促进作用。

Influence of LBP on Expression of BMP-2 during the Process of Fracture Healing in Rats

Abstract:Objective To explore the Influence of lyceum barbarum polysaccharide(LBP) on the expression of BMP-2 during the process of fracture healing in SD rats.To investigate the possibility of LBP used in fracture treatment.Methods 90 3-month-old male SD rats were divided randomly into three groups: LBP high-dose group(H: 160mg·kg-1·d-1),LBP low dose group(L: 40mg·kg-1·d-1) and distilled water group(W)(n=30).All rats were prepared right femoral shaft fracture model with internal fixation with Kirschner wire and were given the drug intervention after operative.The expression of BMP-2 in bone callus were determined in 10 rats of each group at the 2nd,4th,6th week of postoperation by immunohitochemistry.Results The BMP-2 expression of each group continue dto rise at the 2nd,4th,6th week after fracture.The H group expression intensity were significantly higher than that in the L group and W group at each time point.MOD values of BMP-2 expression of H group were higher than that of the L group and W group at each time point(P<0.05).Control observed with HE staining: the cell types of Express BMP-2 positive cells was the same each group.With the development of the fracture healing process,BMP-2 in the fibroblasts,chondrocytes and bone cells within trabecular bone decreased gradually and strong expression was found in the osteoblasts.Tissue sections by HE staining showed: the histology evolvement of H group was earlier than that of L group and W group.Conclusion LBP can promote the expression of BMP-2 on the fracture healing process and this effect shows dose-effect relationship.LBP may have facilitation to the fracture.
