标本采集管与保存方式对超氧化物歧化酶测定结果的影响

摘    要:目的:探讨标本采集管与保存方式对超氧化物歧化酶(SOD)测定结果的影响。方法:随机选取湖北科技学院附二医院80例体检者,前40例体检者同时分别用分离胶促凝真空管(标记为A组)和无抗凝真空管(标记为B组)采集血液标本,离心分离血清,两组都放室温,A组分别于0、4、6、8、10 h测定SOD活性,B组于0 h检测SOD活性;后40例体检者用分离胶促凝真空管(标记为C组)采集血液标本,离心分离血清,置于2-8℃冰箱下保存,分别于0、10、14、21 h测定SOD活性。采用配对t检验分析各组结果差异性。结果:分离胶促凝真空管和无抗凝真空管血清的SOD比较(0 h),差异无统计学意义(P〉0.05);室温下保存的分离胶促凝真空管血清(A组)的SOD于4、6、8、10 h检测,与0 h比较差异具有统计学意义(P〈0.01),与0 h比较相对偏倚分别为3.719%、5.341%、6.807%、8.306%;2-8℃冰箱保存的分离胶促凝真空管血清(C组)的SOD于10、14、21 h检测,与0 h比较差异具有统计学意义(P〈0.01),与0 h比较相对偏倚分别为3.213%、4.552%、5.816%。结论:可采用分离胶促凝真空管采集标本;室温下放置10 h及2-8℃冰箱放置21 h对结果的影响均在临床可接受范围内,与产品说明书上要求的时间相比,室温与冰箱中放置时间都能延长1倍以上。

Abstract:Objective:To explore the influence of specimen collection tube and preservative way on the determination results of superoxide dismutase( SOD). Methods: 80 subjects were randomly selected from the Second Affiliated Hospital,Hubei University of Science and Technology. Firstly,40 subjects blood specimens were simultaneously put in separation gel coagulant vacuum tubes(marked as A group)and in no anticoagulation vacuum tubes(marked as B group). Secondly,40 subjects blood specimens were put in separation gel coagulant vacuum tubes(marked as C group). After centrifugal separation,A group serum was preserved at room temperature,then the SOD was detected respectively at 0 h,4 h,6 h,8 h and 10 h. B group serum was preserved at room temperature,then the SOD was detected at 0 h. C group serum was preserved at 2-8 ℃ refrigerator,then the SOD was detected respectively at 0 h,10 h,14 h and 21 h. Results: The differences of the determination of SOD between separation gel and no anticoagulation vacuum tubes had no statistically significant( P 0. 05). But the determination results of SOD,which were detected in separation gel coagulant vacuum tubes after 4 h,6 h,8 h,10 h at room temperature,were statistically significantly different from the ones detected at 0 h( P 0. 01),and the relative bias of 4 h,6 h,8 h,10 h were 3. 719%,5. 341%,6. 807%,8. 306% respectively in comparison with 0 h. So were specimens in no anticoagulation vacuum tubes and preserved at 2-8 ℃ refrigerator,the relative bias of 10 h,14 h,21 h were3. 213%,4. 552%,5. 816% respectively in comparison with 0 h. Conclusion: Firstly,it is reasonable to use separation gel coagulant vacuum tubes for the specimen collection. Secondly,the determination results of specimens in separation gel coagulant vacuum tubes,both the ones after 10 h at room temperature and the ones after 21 h at2-8 ℃ refrigerator are in the clinical acceptable range. The preservative time can be prolonged by more than 1times compared with the required time in the product specification.
