摘    要:目的探讨戊四氮诱导发育鼠癫癎持续状态(SE)后海马结构的突触重建以及NMDA受体在其中的作用。方法利用图像分析仪测定戊四氮诱导的发育鼠癫癎持续状态模型及用NMDA受体拈抗剂MK-801治疗后海马结构内CA3区及齿状回内分子层突触素(p38)的阳性免疫反应产物的光密度值(A)。结果不同日龄SD大鼠SE后海马结构内各部p38阳性免疫反应产物光密度值较对照组显著增加,尤以CA3区苔藓纤维层和齿状回分子层内带为甚;MK-801可显著减少p38的表达。结论癫癎持续状态后存在突触重建现象,一方面是癫癎发作的结果,另一方面也可能是导致癫癎反复发作的分子学基础;在此过程中NMDA受体发挥着重要的作用。

Abstract:Objective To study the synaptic reorganization of hippocampal formation after pentylenetetrazol-induced status epilepticus in developing rats and the effect of NMDA receptor antagonist MK-801 on it. Methods The optical density(A) of synaptophysin(p38) immunoreactive product was examined by image pattern analysis in the hippocampus of developing rats associated with pentylenetetrazol-induced status epilepticus and MK-801 treatment. Results The A of p38 positive immunoreactive product in hippocmpal formation in the epileptic group was higher than that in the control,especially in the mossy fiber layer of the CA3 and the inner molecular layer of the dentate gyrus.The A in MK-801 group was lower significantly than in epileptic group. Conclusion The increase of p38 expression(related to synaptic reorganization) may be the result from the status epilepticus,as well as the molecular basis of the repeated seizures.NMDA receptors play an important role in the synaptic reorganization.
