地塞米松诱导胚鼠腭突细胞凋亡的形态学观察

摘    要:目的:本研究旨在了解地塞米松(DEX)能否诱导胚鼠腭突细胞调亡,探讨地塞米松致腭裂畸形的机理。方法通过凋亡细胞DNA碎片原位末端标记(ISEL),用光镜、电镜观察10~50mg/kg剂量DEX作用下,15.5孕期日胚鼠腭突变细胞形态变化并作凋亡细胞计数。结果:实验组胚鼠腭突细胞在ISEL法光镜及电镜下观察均存在凋亡的特征性变化;凋亡细胞计数与DEX给药剂量呈正相关(r=0.9911)。结论:DEX可升高胚鼠腭发育过程中的细胞凋亡水平。

Abstract:Objective: The present study was designed to determine whether apoptosis in palatal processes of mouse embryo can be induced by dexamethasone (DEX), so as to provide a new point for explaining DEX how to induce cleft palate. Methods: The morphological changes of cells in palatal processes treated with 10 ̄50mg/kg dose of DEX were observed under light microscrope after in situ end labeling(ISEL) and under electronmicroscope, and apoptotic cells were sumed up as well. Results: In treatment groups, the morphological changes were characterized by some typical features of apoptosis. The sum of apoptotic cells has positive correlation with the doses of DEX (r=0.9911). Conclusions: The apoptosis level in devoloping palate of embryo mouse can be increased by DEX.
