摘    要:目的探讨FAK在大肠上皮不同病变中的表达及临床病理参数的相关性.方法应用免疫组织化学SABC法,分析49例结直肠癌、23例腺癌、10例非肿瘤型息肉.结果49例大肠癌中FAK表达分别是34例(84.4%),FAK的高表达与大肠癌的Duke's分期、浆膜浸润、淋巴结转移、均呈正相关.23例腺癌标本中,FAK表达水平相一致.结论大肠癌的侵袭转移与相关基因协同作用密切相关,在大肠癌的侵袭转移中,FAK的表达具有正负调节的协同作用可能,FAK可为预测大肠癌侵袭转移潜能新的生物学指标.

Abstract:Objective: To detect the role of the FAK expression in different human colonic lesions and normal tissues. Further, to get the correlation between expression of FAK and clinicopathological data including tumor differentiation, Duke's stage etc. Methods: Using SP immunohistochemical techniques combined with imaging,the expression of FAK was analysed in 49 colonic Carcinoma, 23 adenoma and 10 polypoid lesions. Results: FAK was weakly expression in most benign colonic epithelium but was up-regulated at moderate or strong levels in invasive colon carcinoma. Immunohistochemical analyses revealed a correlation between tumor stage and the levels of FAK, especially positively expressed on vascular endothelium and lymphatic endothelium of adjacent infiltration carcinoma. The staining of FAK was higher in cancer of low differentiation cell than of well differentiation. The cancer with metastasis had more FAK protein than the cancers without metastasis. FAK expression levels were significantly related to Duke's stage. FAK expression levels were not related to tumor size. Conclusion: FAK is significantly overexpressed in colon carcinoma, implying that FAK may play an important role in colon carcinomas or colon carcinoma progression. FAK protein expression level in colon cancer tissues may serve as a new parameter for judging tumor biological behavior and judging prognosis.
