双向分化肿瘤血管生成拟态的分子机制初步观察

摘    要:目的 研究肿瘤细胞分泌IV型胶原和过碘酸雪夫氏反应 (PAS)阳性物质在血管生成拟态 (VM)中的作用 ,以及VM对肿瘤细胞表达血管内皮细胞生长因子 (VEGF)的影响。方法 收集双向分化恶性肿瘤石蜡包埋样本 158例 ,制作成组织芯片 ,进行CD3 1和VEGF双重染色及IV型胶原染色。通过网格计数法 ,比较CD3 1与PAS阳性图案围成的管道面积差别及分布特点 ,以及有VM的点阵和无VM的点阵VEGF表达的差别。结果 VM的基底膜样结构表现为PAS及IV型胶原阳性 ;无VM的双向分化肿瘤VEGF表达高于有VM者 ,其中在恶性黑色素瘤及腺泡型横纹肌肉瘤中 ,二者VEGF的表达差异有显著性 (P <0 .0 5)。结论 PAS阳性物质和IV型胶原参与构建了VM的管壁 ,VEGF表达的差别表明VM的存在可以提供肿瘤细胞氧气和营养物质

Abstract:Objective To study the relationship between collgen (Col) IV, VEGF secreted by the tumor cells and vasculogenic mimicry (VM). Methods 158 bi-phas e differential malignant tumor specimens were alloted and made into tissue micro array. These tissue microarray sections were stained with CD31, periodic acid-S chiff (PAS) and Col IV. Subsequently, distributive trait of Col IV and the diffe r ence of VEGF expression were analyzed. Results The basement membrane of VM was PA S and Col IV positive. The expression of VEGF in bi-phase differential ma lignant tumor with VM was less than that in those without VM. The difference of VEGF ex pression in malignant melanoma and alveolar rhabdomyosarcoma was significant ( P<0.05). Conclusion Collgen IV and periodic acid-Schiff positive material take part in constructi ng the basement membrane of vasculogenic mimicry. The difference of the VEGF exp ression proves that vasculogenic mimicry can sustain the tumor blood supply.
