低剂量辐射对小鼠外周血干/祖细胞的动员作用

摘    要:目的:探讨低剂量辐射对小鼠外周血干/祖细胞的动员效应。方法:采用深部X线对小鼠进行低剂量辐射,甲基纤维素半固体培养、流式细胞仪检测单纯低剂量辐射及联合粒细胞集落刺激因子(G-CSF)后外周血粒单系造血祖细胞集落(CFU-GM)及造血干祖细胞数量(c-kit+细胞数)。结果:小鼠受到25~75 mGy照射后外周血CFU-GM集落数明显增多,c-kit+细胞数平行增加,其中以75 mGy组最明显;受照后24 h CFU-GM集落数开始增加,72 h最明显,96 h后有所下降。75 mGy+半量G-CSF组周围血CFU-GM集落数及c-kit值均明显高于单纯低剂量照射组,接近全量G-CSF组。结论:低剂量辐射对小鼠外周血干/祖细胞有动员效应,联合应用G-CSF有明显的协同作用。

Abstract:Objective To study the mobilizing effect induced by low dose irradiation on mouse peripheral blood(stem/progenitor) cells.Methods Peripheral blood CFU-GM was cultivated in methylcellulose semi-solid culture system,and c-kit~ cells were counted by flow cytometry after low dose irradiation or combined with G-CSF.(Results The) CFU-GM and c-kit~ cells of peripheral blood in irradiated mice were much higher than those in control mice,the CFU-GM started to increase at 24 h after irradiation,reached maximum at 72 h and stayed high level till 96 h.The CFU-GM and c-kit~ cells at 72 h after 75 mGy irradiation were the most.The CFU-GM and c-kit~ cells in semi-dose G-CSF 75 mGy group were much higher than those in simple low dose irradiation group and were close to those in the group dealed with adequate G-CSF.Conclusion Low dose irradiation can mobilize peripheral blood stem/progenitor cells,and low dose irradiation combined with G-CSF can produce synergetic effect.
