白细胞介素1受体拮抗剂对大鼠局灶性脑缺血再灌注神经细胞凋亡的影响

摘    要:目的 探讨白细胞介素1受体拮抗剂(IL-1ra)对大鼠局灶性脑缺血再灌注神经细胞凋亡的影响。方法 取成年雄性SD大鼠24只,随机分为三组,每组8只:假手术组(Sham组)、局灶性脑缺血组(MCAO组)和脑缺血+ IL-1ra干预组。术后24 h断头取脑, 应用TUNEL染色法,检测梗死灶周围神经细胞凋亡情况,进行计数并进行统计学处理。结果 与假手术组相比,局灶性脑缺血组神经细胞凋亡显著增多(P<0.01); 与局灶性脑缺血组相比,IL-1ra干预组神经细胞凋亡显著减少(P<0.01)。结论 IL-1ra可抑制大鼠脑缺血梗死灶周围神经细胞凋亡.

Abstract:Abstract Objective To study the effects of IL-1 receptor antagonist(IL-1ra)on neurocyte apoptosis in cerebral ischemia in rats. Methods 24 male SD rats were randomly assigned into 3 groups: sham operation group, middle cerebral artery occlusion (MCAO) group and MCAO+IL-1ra treatment group. Terminal deoxynucleotidyl transferase-mediated biotinylated UTP nick end labeling (TUNEL) staining was used to detect in situ cell apoptosis. Results As compared with sham operation group, cerebral ischemia significantly induced neurocyte apoptosis (P<0.01). However, neurocyte apoptosis in IL-1ra-treated group was markedly decreased in comparison with MCAO group (P<0.01). Conclusion IL-1ra can suppress neurocyte apoptosis in cerebral ischemia.
