成人腮腺及其胚胎发生的免疫组织化学研究

摘    要:目的:研究成人腮腺的胚胎发生。方法;18例胚胎腮腺和6例成人腮腺HE、PAS染色进行形态学观察。免疫组化ABC法观察抗角蛋白,波形蛋白,胶原纤维酸性蛋白,肌动蛋白在组织中的表达。结果;15周的胚胎腮腺可见实性细胞团,17周时分支增多,20周时出现小叶轮廓,28周形态发育基本成熟。胚胎腮腺中4种蛋白均有表达,成人腮腺中角蛋白,胶原纤维酸性蛋白在导管有表达,肌动蛋白仅标记肌上皮,波形蛋白为阴性。结论

Abstract:Objective: To study the embryonic development of adult parotid gland. Methods: Using HE and PAS staining, as well as immunohistochemical staining with monoclonal antibodies against cytokeratin, vimentin, glial fibrillary acidic protien (GFAP) and smooth muscle actin, we studied embryonic and adult parotid glands (18 and 6 cases respectively). Results: For the embryonic parotid glands, we found solid cell masses at 15th week of embryo. Branches increasing at 17th week; Outline of lobules at 20th week. At 28th week, the glands became almost mature. All of the four proteins were positive in embryonic parotid glands. But in adult parotid glands Keratin and GFAP were positive in the cells of ducts, actin was detected in myoepithelial cell only. Vimentin are negative for all cells of adult parotid gland.Conclusions: Morphogenesis is accompanied by the functional differentiation of the parotid gland cells. The embryonic cells of parotid gland have multiple differential potential. The peripheral cells of terminal tubules in embryonic parotid glands are active. Maybe they differential to part of the basal cells of excretory duct and myoepithelial cells in adult parotid glands.
