来氟米特联合贝那普利对糖尿病肾病大鼠ICAM-1表达的影响

摘    要:目的 研究来氟米特联合贝那普利对糖尿病肾病大鼠对细胞间粘附分子(ICAM-1)表达的影响.方法 成年雄性Wistar大鼠,腹腔注射枸橼酸钠缓冲液,为正常对照组(NC组),同批大鼠腹腔注射STZ(65 mg/kg),建立糖尿病模型后,随机分为模型组(DN组)、贝那普利组、联合用药组.贝那普利组每天给予贝那普利灌胃(10 mg/kg/d),联合用药组每天给予来氟米特和贝那普利灌胃(分别灌10 mg/kg/d),NC和DN组每天给予等量蒸馏水灌胃.第4周、8周、12周末检测24小时尿蛋白排泄量(24UPE)和尿肌酐(Ucr)、血糖(Glu)、血肌酐(Scr)、计算内生肌酐清除率(Ccr),12周末观察肾脏组织病理学变化及肾脏组织中ICAM-1的表达水平.结果 DN对照组ICAM-1在肾小球系膜区有强烈表达(P<0.01),贝那普利组较DN组表达减轻,联合用药组较贝那普利组表达更明显减轻(P<0.05).结论 来氟米特联合贝那普利可显著减少糖尿病肾病大鼠肾组织内ICAM-1表达,从而延缓糖尿病肾病的进展,其效果优于单用贝那普利.

Effect of combination of benazepril and leflunomide on ICAM-1 expression in the renal tissues of diabetic nephropathy rats

Abstract:Objective To investigate the effect of combination of benazepril and leflunomide on expression of inter- cellular adhesion molecule (ICAM-1) in diabetic nephropathy (DN) model rats. Methods fifty for male Wistar rats were randomly divided into five groups., normal control (NC) group,DN group (DN), benazepril group and combina- tion (benazepril + leflunomide group. Diabetic Nephropathy models were induced by Intraperitoneal injection with Streptozotocin (STZ, 65 mg/kg) in the groups except NC. Benazepril group was intragastric administered with benaze- pril (10 mg/kg/d) and combination group was intragastric administered with leflunomide and benazepril (10 mg/kg/d, respectively), Same volume of water was intragastric administered in the other groups. At the end of 4th, 8th and, 12th weeks,24-bour urinary protein excretion (24UPE) and urine creatinine (Ucr),blood glucose level, serum creatinine (Scr) and endogenous creatinine clearance rate calculation (Ccr) were examined. At the end of 12th weeks,the patho- logical changes and expression of ICAM-1 were detected in the renal tissue. Results In DN group,ICAM-1 expression was significantly increased in mesangial region of glomerulus (P〈0.01 ,compared with NC group) ; the increases were attenuation in combination groups expression (P〈0.05, respectively), and the" attenuation of combination group was larger than benazepril group(P〈0.05). Conclusion Combination of leflunomide and benazepril can significantly re- duce the ICAM-1 expression in renal tissue in DN rats and can delay the progress of DN, and the effect is superior to benazepril alone.
