miR-143和miR-520d-5p在胃癌组织中的异常表达

摘    要:目的 应用MicroRNAs (miRNAs)芯片技术筛选胃癌组织中差异表达的miRNAs,研究与胃癌相关的miRNAs.方法 从3对手术切除的胃和相配对的正常胃组织中提取总RNA,miRNA芯片检测miRNA表达情况,然后在47例配对的胃癌和正常胃组织中,用Real-time RT-PCR(Taqman)对部分差异表达miRNA进行验证.结果 芯片结果显示:与配对的正常胃组织相比,有62个miRNAs在胃癌中异常表达,其中42个miRNA高表达,20个miRNA低表达.用Real-time RT-PCR(Taqman)验证显示:miR-143在胃癌组织中表达显著降低(JP=0.002 9);miR-520d-5p在胃癌组织中表达显著提高(P =0.032).结论 miR-143和miR-520d-5p可能参与了胃癌的发生过程.

Abstract:Objective To study the expression profiles of miRNAs in gastric cancer by miRNAs microarray and the relationship between miRNAs and gastric cancer. Methods Cancer tissues and adjacent noncancerous tissues were surgically reseeted, and RNA was extracted by Trizol reagent, miRNA expression profiles in 3 pairs of gastric cancer and adjacent nontumorous tissues were analyzed using a mammalian miRNA microarray. Aberrant expression of miRNAs was further verified by Real-time PCR in 47 pairs of gastric cancer and adjacent nontumorous tissues. Results 63 aberrant miRNAs were identified by miRNA microarray, including high expression of 42 miRNAs and low expression of 21 miRNAs. Meantime, Real-time PCR determined that the expression of miR-143 was decreased and miR-520d-Sp was increased in cancer tissues compared to normal tissues. Conclusion miR-143 and miR-520d-Sp may be involved in the process of gastric carcinogenesis.
