Abstract:Objective To investigate the effects of electroacupuncture of pericardium meridians acupoints on SYP and PSD-95 in serum and brain tissue of MCAO rats;To explore the mechanism of acupuncture on synaptic plasticity after cerebral ischemia.Methods Ninety SD rats were randomly divided into normal group,sham-operation group,model group,pericardial meridians group and lung meridians group.Each group was divided into three time points of 14,21 and 28 days,with six rats in each time point.The model of focal cerebral ischemia was established by inserting thread into external carotid artery.“Tianquan”,“Quze”,“Neiguan”and“Daling”acupoints were selected by pericardial meridiansgroup.In the lung meridians group,“Tianfu”,“Chize”,“Lieque”and“Taiyuan”acupoints were selected,and electroacupuncture treatment was started on the second day after the model was established successfully,30 minutes each time,6 days in a row,1 day apart.The contents of SYP and PSD-95 in serum were detected by ELISA,and the expressions of SYP mRNA and PSD-95 mRNA in infarcted brain tissue were detected by RT-qPCR.Results Compared with the normal group and the sham-operation group,the serum levels of SYP and PSD-95 in 21d and 28 d(P<0.05,P<0.01)in model group decreased significantly,and the expressions of SYP mRNA and PSD-95 mRNA in the brain tissue of the model group decreased at each time point(P<0.01).Compared with the model group,the serum SYP in the 21 d pericardial meridians group increased significantly(P<0.01),28 d serum PSD-95 increased significantly(P<0.05),and the expressions of SYP mRNA and PSD-95 mRNA increased at all time points(P<0.01).Compared with at the same time point,the expressions of SYP and PSD-95 in the serum of the pericardial meridians group and the expressions of SYP mRNA and PSD-95 mRNA in the brain tissue were superior to the lung meridians group(P<0.01).Conclusion Electroacupuncture of pericardial meridians can improve the content of SYP and PSD-95 in serum and SYP mRNA and PSD-95 mRNA expressions in brain tissue,and regulate the plasticity of synapse after cerebral ischemia.
