福辛普利和普伐他丁对糖尿病大鼠肾脏细胞凋亡的影响

摘    要:目的 探讨血管紧张素转换酶抑制剂 (ACEI)和羟基戊乙酰辅酶A(HMG CoA)还原酶抑制剂对糖尿病大鼠肾脏功能的影响及其机制。方法 腹腔内注射链脲佐菌素 (STZ) ,诱发大鼠糖尿病模型 ,每日给予福辛普利 15mg/(g·d)或 /和普伐他丁 2 0mg/(kg·d)灌胃 ,共 4周。采用末端原位标记法检测肾脏细胞凋亡情况 ,并观察血糖、血脂、尿素氮、内生肌酐清除率等相关生化指标。结果 糖尿病组较对照组肾脏凋亡细胞明显增多 ,福辛普利治疗组及联合治疗组凋亡细胞明显减少 (P <0 .0 1)。结论 福辛普利和普伐他丁可能通过抑制细胞凋亡而发挥肾功能的保护作用

Abstract:Objective To investigate the effects of angiotensin converting enzyme inhibitor (ACEI) and HMG CoA reductase enzyme inhibitor on apoptosis in the kidney of diabetic rats.Methods Diabetes was induced by intraperitoneal injection of STZ (50 mg/kg). Four weeks later, Fosinopril (10 mg/kg every day) or Paravasatin (20 mg/kg every day) was delivered daily by gavage for 4 weeks. Apoptosis in renal cortex was detected by means of terminal deoxynucleotidyl transferase mediated d UTP nick end labeling (TUNEL). Blood glucose, blood lipid, BUN anc Ccr were measured.Results Compared with those in control group, apoptotic cells were significantly increased in the diabetic group ( P < 0.01 ). In the Fosinopril treated group or combine treated group, the apoptotic cells were significantly decreased ( P < 0.01 ).Conclusion The renal protective effect of Fosinopril and pravastatin may be partly through inhibiting apoptosis.
