摘 要: | 目的:研究逍生散颗粒剂对干眼小鼠模型泪腺Bax mRNA、Bcl-2 mRNA表达的影响。方法选择c57小鼠,分为空白对照组、干眼模型组、逍生散低剂量组、逍生散中剂量组、逍生散高剂量组,测定泪液分泌量和泪膜破裂时间,采用荧光定量PCR的方法检测泪腺中Bax、Bcl-2的mRNA含量。结果(1)干眼模型组的泪液分泌试验Ⅰ( SIT)、泪膜破裂时间( BUT)低于空白对照组,逍生散组的SIT、BUT均高于干眼模型组,且逍生散中剂量组的SIT、BUT均高于逍生散低剂量组和逍生散高剂量组;(2)干眼模型组泪腺组织Bcl-2的mRNA含量低于空白对照组、Bax的mRNA含量高于空白对照组,逍生散低剂量组、逍生散中剂量组、逍生散高剂量组泪腺组织Bcl-2的mRNA含量高于干眼模型组、Bax的mRNA含量低于干眼模型组;(3) SIT、BUT与泪腺中Bax的mRNA含量呈负相关、Bcl-2的mRNA含量呈正相关。结论逍生散颗粒剂能够调节干眼小鼠模型泪腺中Bcl-2、Bax的mRNA,3.6 mg/g·d中等剂量的调节作用最为明显,且Bcl-2、Bax的含量与干眼的体征存在相关性。

Abstract: | Objective To study the effect of Xiaosheng San Granules on Bax mRNA, Bcl-2 mR-NA expression in lacrimal gland of dry eye mouse model. Methods c-57 mice were enrolled and randomly divided into blank control group, dry eye model group, XiaoSheng San low dose group, XiaoSheng San middle dose group, XiaoSheng San high dose group. Then tear secretion and tear break-up time were detec-ted and Bax mRNA, Bcl-2 mRNA expression in lacrimal gland detected by fluorescence quantitative PCR. Results (1) SIT, BUT of dry eye model group were lower than those of control group;SIT, BUT of Xi-aoSheng San group were higher than those of dry eye model group;SIT, BUT of XiaoSheng San middle dose group were higher than those of low dose group and high dose group. ( 2 ) Bcl-2 mRNA levels of dry eye model group were lower and Bax mRNA levels were higher than those of control group;Bcl-2 mRNA levels of XiaoSheng San low dose group, XiaoSheng San middle dose group, XiaoSheng San high dose group were higher and Bax mRNA levels were lower than those of dry eye model group;(3) SIT, BUT were negatively correlated with Bax mRNA levels in lacrimal gland and positively correlated with Bcl-2 mRNA levels. Con-clusion Xiaosheng San Granules can regulate Bax mRNA, Bcl-2 mRNA expression in lacrimal gland of dry eye mouse model and regulating effect of 3. 6 mg/g·d middle dose is the most obvious; the contents are correlated with dry eye symptoms. |
