硒、锌对索曼神经毒剂诱导大鼠过氧化损伤的保护机制

摘    要:目的:研究索曼对大鼠急性中毒的自由基损伤作用及硒、锌的抗氧化保护作用。 方法:雄性大鼠40只,随机分为阴性组(正常对照组)、阳性组(损伤对照组)、硒组(加硒保护组)和锌组(加锌保护组)。测定指标有血清、大脑、肝脏的维生素E、总抗氧化能力(total antioxidant capacity,T-AOC)和一氧化氮合成酶(nitric oxide synthase,NOS)。 结果:索曼中毒后,大鼠大脑与肝脏中的T-AOC分别从(80.60±9.87),(174.63±6.67)mmol/g降至(38.69±3.57),(97.40±5.97)mmol/g(t=16.22,14.8,P<0.01);维生素E含量从(3.78±0.49),(37.40±3.36)μmol/g降至(1.52±0.38),(17.94±2.75)μmol/g(t=13.91,13.50,P<0.01),血清中的T-AOC和维生素E也有所降低(t=2.31,2.85,P<0.05);而血清NOS活性从(6.70±0.30)mmol/L升高至(15.60±2.78)mmol/L(t=14.92,P<0.01),大脑与肝脏中的NOS活性也有所升高;硒、锌组较阳性对照组损伤指标变化幅度较小。 结论:索曼中毒有明显的自由基损伤作用,硒、锌对索曼中毒有显著的抗氧化保护作用。

Abstract:AIM:To study the action of free radical injury in soman acute intoxication and the protective effect of Se and Zn on lipid peroxidative injury induced by soman. METHODS:Forty male rats were randomly divided into 4 groups:control group,soman group,Se group,Zn group.Vitamin E(VE) content,nitric oxide synthase(NOS) activity and total antioxidant capacity(T-AOC) in serum, cerebrum and liver of soman-poisoned rats were determined respectively. RESULTS:After soman intoxication, in cerebrum and liver,T-AOC activity decreased from(80.6± 9.9),(174.6± 6.7) mmol/g protein to(38.7± 3.6),(97.4± 6.0) mmol/g(t=16.22,14.8,P< 0.01),respectively; VE content decreased from(3.8± 0.5),(37.4± 3.4) μ mol/g protein to (1.5± 0.4),(18.0± 2.8) μ mol/g(t=13.91,13.50,P< 0.01),respectively.In serum,VE content and T-AOC activity also decreased(t=2.31,2.85,P< 0.05);But in serum,NOS activity increased from(6.7± 0.3) mmol/L to(15.6± 2.8) mmol/L(t=14.92,P< 0.01).In cerebrum and liver,NOS activity also increased;compared with soman group,the changes were less significant in Se and Zn group. CONCLUSION:There is a significant free radical injury induced by soman in rats,Se and Zn are protective against the injury.
