滋补肝肾方及其拆方对过氧化氢损伤小鼠B16黑素细胞的影响

摘    要:目的:观察滋补肝肾方及其拆方对过氧化氢损伤小鼠B16黑素细胞的保护作用,探讨其组方配伍的作用基础。方法:实验分为6组:①正常对照组;②模型组;③祛风组;④和血组;⑤补肝肾组;⑥全方组。将10%各组血清预先与B16细胞孵育24h,再以1mmol/L H2O2作用于细胞0.5h。采用MTT法检测各组血清对B16细胞活性的影响,采用生化法测定细胞内超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活力,丙二醛(MDA)含量。结果:经H2O2刺激后B16细胞活性显著下降(P0.01)。与模型组相比,各组含药血清可提高细胞活性,能不同程度地提高SOD、CAT、GSH-Px活性(P0.05,P0.01),同时降低MDA含量和羟自由基(OH-)的产生(P0.05,P0.01)且全方组较优。结论:滋补肝肾全方及拆方均可减轻过氧化氢诱导B16细胞的氧化损伤,全方组的效果最好,体现了中医治疗白癜风的滋补肝肾、养血祛风综合作用的科学性。

Abstract:Objective: To observe the protective effect of Zibuganshenfang and its components conditioning against H2O2 induced Mice B16 Melanocytes damage and investigate the possible mechanism.Methods: B16 Melanocytes were divided into 6 groups: ①Normal group(N);②Model group(M);③Qufeng group(QF);④Hexue group(HX);⑤Buganshen group(BGS);⑥zibuganshenfang group(ZBGSF).B16 Melanocytes were stimulated with 1mmol/L H2O2 for 0.5h after pre-treatment with 10% different serum for 24h.MTT method were adopted to determine cellular activity.The activity of SOD、CAT、 GSH-Px,MDA content and OH-were detected by biochemical methods.Results: B16 cell vitality decreased after H2O2 stimulating(P<0.01).Treatment groups improved the cell survival activities,increased SOD,CAT and GSH-Px activity(P<0.05,P<0.01),reduced OH-releasing and MDA content(P<0.05,P<0.01).Among of all the index,the curative effect of ZBGSF group was the best.Conclusion: ZBGSF and all of its components can protect B16 cell from oxidative damage by H2O2,and the curative effect of ZBGSF is best.This study proves the rationality of Zibuganshen method ’s synthetic action in treating vitiligo.
