IgA肾病患者肾组织内电压依赖性Ca^2+通道和高通透性Ca^2+激活钾通道mRNA的表达

摘    要:目的观察IgA肾病(IgAN)患者肾组织内电压依赖性Ca2 通道(VOCC)和高通透性Ca2 激活钾通道(BKCa)mRNA表达情况,以评价VOCC和BKCa通道mRNA表达与IgAN患者肾脏病理损害的相关性。方法提取对照组(n=11)和IgAN患者(n=25)肾组织总RNA,以RT-PCR方法检测VOCC和BKCa mRNA表达量,同时将IgAN患者肾活检组织的一部分进行病理诊断和病理损害评分,病理损害评分采用Katafuchi半定量法。结果与对照组比较,IgAN患者肾组织内VOCC和BKCa mRNA的表达均呈显著增高(P<0.05);IgAN患者肾组织内VOCC mRNA表达与病理损害总积分、肾小球系膜细胞增生程度均呈正相关(r=0.6962,P<0.01;r=0.4220,P<0.05),而与肾小球系膜基质增多和肾小球硬化程度无相关性(P>0.05);IgAN患者肾组织内BKCa mRNA表达与病理损害总积分呈正相关(r=0.5582,P<0.05),而与肾小球系膜细胞增生程度、肾小球系膜基质增多以及肾小球硬化程度均无相关性(均P>0.05)。结论IgAN患者肾组织内VOCC和BKCa mRNA表达异常,两种通道蛋白的mRNA表达异常与肾小球组织病理损害总积分呈一定程度正相关,提示IgAN患者肾组织内VOCC和BKCa的表达情况,可作为IgA肾病进展与否的指标。

VOCC and BKCa mRNA expression in kidney tissues of IgA nephropathy patients

Abstract:Objective To explore the mRNA expression of voltage-operated Ca2 channel(VOCC) and large-conductance Ca2 -activated K channel(BKCa) in kidney tissues of IgA nephropathy patients,and investigate the correlations between the expression of VOCC mRNA and BKCa mRNA and the degree of renal pathologic lesions. Methods The total RNA was extracted from kidney tissues of 11 normal controls and 25 IgA nephropathy patients for the detection of the expression of VOCC mRNA and BKCa mRNA by RT-PCR.A part of the kidney tissues of IgA nephropathy patients were subjected to pathologic diagnosis,and the pathological lesions integrals of glomerulus was evaluated referring to Katafuchi methods.Results The expression of VOCC mRNA and BKCa mRNA in kidney tissues of IgA nephropathy patients significantly increased compared to those of controls(P<0.05).There was positive correlation between the abnormal expression of VOCC mRNA and BKCa mRNA and total glomerular pathological lesions integrals(r=0.6962,P<0.01 for VOCC mRNA expression,and r=0.5582,P<0.05 for BKCa mRNA expression).There was positive correlation between the abnormal expression of VOCC mRNA and mesangial cell proliferation integrals(r=0.4220,P<0.05).There was no correlation between the abnormal expression of VOCC mRNA and mesangial extracellular matrix integrals and glomerular sclerosis integrals(P>0.05),there was no correlation between the abnormal expression of BKCa mRNA and mesangial cell proliferation integrals,mesangial extracellular matrix integrals and glomerular sclerosis integrals(P>0.05). ConclusionThe expression of VOCC mRNA and BKCa mRNA in kidney tissues of IgA nephropathy patients are abnormal.There is positive correlation between the abnormal expression of VOCC mRNA and BKCa mRNA and total glomerular pathological lesions integrals.The expression of VOCC mRNA and BKCa mRNA in kidney tissues of IgA nephropathy may serve as the indictor for the disease progression.
