高同型半胱氨酸血症家兔模型的建立和叶酸的预防治疗作用的实验研究

目的　建立家兔的高同型半胱氨酸血症 (hyperhomocysteinmia)模型 ,观察该血症对动脉损伤以及叶酸对该血症的影响。方法　给家兔皮内注射同型半胱氨酸 (homocysteine,Hcy) 2 0 mg·kg-1,间隔 1 2 h注射一次。结果　皮内注射同型半胱氨酸可使家兔血中同型半胱氨酸浓度明显增高 ,连续注射 3 0 d,可使家兔动脉内膜内皮细胞增生 ,中膜见点状或小灶状脂质浸润 ,V.G.弹力纤维染色示不同程度的弹力纤维断裂伸直 ,异染性物质呈阳性反应。皮内注射同型半胱氨酸前经口灌胃给予复方叶酸片 (叶酸 0 .0 4 mg· kg-1,Vit B60 .5 mg·kg-1,Vit B12 5 .75μg· kg-1)和叶酸片 (0 .0 4 mg· kg-1)均可预防家兔血中同型半胱氨酸浓度升高 ;皮内注射同型半胱氨酸 2 h后经口灌胃给予复方叶酸片和叶酸片也可使血中升高了的同型半胱氨酸浓度降低 ,但复方叶酸片防治高同型半胱氨酸血症的效果比单一叶酸好。复方叶酸片和叶酸片均可减轻家兔血中高浓度同型半胱氨酸对血管的伤害。结论　皮内注射同型半胱氨酸 2 0 mg· kg-1,间隔 1 2 h一次 ,可致家兔高同型半胱氨酸血症 ,最终致家兔动脉粥样硬化 ;复方叶酸片和叶酸片均可预防和治疗高同型半胱氨酸血症 ;并减轻同型半胱氨酸对动脉的损害

Establish the model hyperhomocysteimia with rabbits and research the prophylactic and therapensis function of the folic

Objective The hyperhomocysteinmia rabbit model was established, and the injurious effect of it on artery and the prevented function of the folic acid was investigated. Method Rabbits were given homocysteine(20 mg·kg -1) intradermally every 12 hours. Results The concentration of homocysteine in blood increased significantly in model rabbits. After 30 days, in model group, the endothelia cell hyperplasia on intima can be observed, tunica media was infiltrated by the dot or local lipid, and elastic fibers had different rupture level with the different V.G staining. The elevation of the blood concentration of homocysteine was inhibited by administration the compound folic tablet (include folic 0.04 mg·kg -1, VitB 60.5 mg·kg -1, VitB 125.75 μg·kg -1) and the folic tablet(0.04 mg·kg -1) before or after homocysteine, but the effect of the compound folic was better than that of the single folic. Conclusion The model of hyperhomocysteinmia can be established after being given homocysteine subcutaneously every 12 hours. The compound folic tablet and folic tablet both prevent and therapy the hyperhomocysteinmia, diminish the artery damage.
