丹参多酚酸HPLC指纹图谱研究

摘    要:目的:建立丹参多酚酸原料的指纹图谱研究方法。方法:以丹酚酸B为参照物,采用高效液相色谱法,色谱柱为Luna C18(2)100A(250mm×4.6mm,5μm),流动相为乙腈-0.026%磷酸水溶液(梯度洗脱),检测波长为286nm,柱温为30℃,流速为1mL.min-1。结果:从所建指纹图谱中确定了5个共有峰,建立了丹参多酚酸的共有模式;在10批丹参多酚酸中有6批样品的指纹图谱相似度在0.90以上。结论:本方法简便、准确、重现性好,可为评价丹参多酚酸原料药的质量提供依据。

Abstract:OBJECTIVE: To establish a method for the study of HPLC fingerprints of salvianolic acids. METHODS: The salvianolic acid B was used as reference substance. The HPLC was employed in which the chromatographic column was Luna C18(2) 100A(250mm×4.6mm,5μm) at a column temperature of 30℃. The mobile phase was a mixture of acetonitrile- 0.026% phosphoric acid water solution(in a gradient elution mode) at a flow rate of 1 mL·min^-1.The detection wavelength was 286 nm. RESULTS: 5 common peaks were established in the chromatographic fingerprints of salvianolie acids, and the mutual model of salvianolic acids has been established. 6 of the total 10 batches of samples had a similarity of above 0.90. CONCLUSION: The method is simple, accurate and reproducible, and it serves as a basis for the quality evaluation of the crude drug of salvianolie acids.
