灯盏花素对大鼠脑缺血再灌注损伤后胶质原纤维酸性蛋白和NOS的影响

摘    要:目的:研究灯盏花素对大鼠脑缺血再灌注(I-R)损伤后胶质原纤维酸性蛋白(GFAP)和一氧化氮合酶(NOS)的影响。方法:实验分为5组,即假手术、I-R、MgSO4及灯盏花素高、低剂量组。采用线栓法复制脑I-R损伤模型大鼠,观察灯盏花素对模型大鼠GFAP和NOS的作用。结果:与假手术组比较,I-R组大鼠GFAP免疫染色呈强阳性,血清中NOS和诱导型NOS(iNOS)活性显著增强(P<0.01)。与I-R组比较,灯盏花素高、低剂量组大鼠GFAP免疫染色则呈阳性-弱阳性改变,灯盏花素高剂量组大鼠iNOS显著降低(P<0.01)。结论:灯盏花素对I-R损伤模型大鼠有保护作用,其机制可能与抑制GFAP和NOS表达等作用有关。

Effect of Breviscapine Injection on the Expression of GFAP and NOS in Rats with Cerebral Ischemia-Reperfusion Injury

Abstract:OBJECTIVE: To study the effect of breviscapine injection on glial fibrillary acidic protein (GFAP) and nitri-coxide synthase (NOS) in rats with isehemical reperfusion(I-R) injury. METHODS: The rats were divided into 5 groups: sham group, I-R group, MgSO4 group, breviscapine low dose group and high dose group. Cerebral I-R injury model was established with suture method.The effect of breviscapine injection on GFAP and NOS were evaluated. RESULTS: In is- chemia- reperfusion group compared with the sham group, the GFAP staining was strongly positive, serum NOS and inducible NOS (iNOS) activity were significantly enhanced(P〈0.01) .In breviscapine - treatment groups(high dose and low dose groups) compared with ischemic- reperfusion group, GFAP staining changed from positive to weak positive, and iNOS was significantly lowered in breviscapine high dose group. CONCLUSION: Breviscapine injection has protective effect on focal cerebral isehemia - reperfusion injury, which might be attributed to its action in inhibiting the expression of GFAP and NOS.
