参芪扶正注射液对心力衰竭模型大鼠心肌MMP-3、TIMP-1的作用研究

摘    要:目的:研究参芪扶正注射液对心力衰竭模型大鼠心肌膜细胞基质金属蛋白酶-3(MMP-3)及其组织抑制剂-1(TI MP-1)的作用。方法:实验分为6组,即正常(等体积生理盐水)、模型(等体积生理盐水)、卡托普利(10mg.kg-1)和参芪扶正注射液高、中、低剂量(6、4、2mL)组。除正常组外,其它大鼠均于实验开始后腹膜内注射阿霉素以复制心力衰竭模型,同时给予相应药物。药物干预8周后,将大鼠麻醉处死,迅速取心脏标本,进行病理检查及用RT-PCR法对大鼠心肌MMP-3、TI MP-1mRNA进行半定量分析。结果:与正常组比较,模型组大鼠心肌MMP-3mRNA显著升高(P<0.01),TI MP-1mRNA显著降低(P<0.01);与模型组比较,参芪扶正注射液中、低剂量组大鼠心肌MMP-3mRNA表达显著降低(P<0.05),TI MP-1mRNA显著升高(P<0.05)。结论:参芪扶正注射液可抑制MMP-3的表达,上调TI MP-1的表达,改善心肌重塑,从而达到抗心力衰竭作用。

Effects of Shenqi Fuzheng Injection on the Expression of MMP - 3 and TIMP - 1 in Rats with Congestive Heart Failure

Abstract:OBJECTIVE: To observe the effects of Shenqi Fuzheng injection on the expression of MMP - 3 and TIMP - 1 in congestive heart failure model rats. METHODS: The experimental rats were randomly divided into 6 groups, i.e. normal group, model group, captopril(10 mg - kg 1) group, Shenqi Fuzheng injection low dosage (2 mL) group, Shenqi Fuzheng injection medium dosage(4 mL)group, and Shenqi Fuzheng injection high dosage (6 mL)group. All except the normal group were intraperitoneally injected with adriamycin on the first experimental day. After drug intervention for 8 weeks, the rats were anesthetized and sacrificed, with heart specimen sampled rapidly for pathological examination and semi -quantitative analysis of myocardial MMP - 3 and TIMP - 1 mRNA by RT - PCR method .RESULTS: In the model group compared with the normal group, MMP 3 mRNA were significantly increased(P〈0.01),TIMP 1 mRNA were significantly decreased(P〈0.01) .Compared with model group, in Shenqi Fuzheng medium dosage injection group and Shenqi Fuzheng low dosages group, the expressions of MMP - 3mRNA were significantly lower(P 〈 0.05) but the expressions of TIMP - 1 mRNA were significantly higher(P 〈 0.05). CONCLUSION: The anti - heart - failure efficacy of Shenqi Fuzheng injection is achieved through down regulating the expression of MMP - 3, up - regulating the expression of TIMP - 1 and improving cardiac remodeling.
