摘    要:目的:观察乙酰胆碱(ACh)对大鼠淋巴细胞由刀豆素A(ConA)诱导的白细胞介素2(IL-2)生成的影响。方法:取大鼠的脾脏制成单个细胞悬液进行体外培养,用ConA诱导脾细胞的IL-2生成,然后以MTT比色法间接测定IL-2的生成。结果:ACh,M型胆碱能受体激动剂毛果芸香碱和N型胆碱能受体激动剂烟碱在10^-10-10^-6mol/L浓度范围都能显著增强ConA诱导的IL-2生成,M型胆碱能受体阻断剂阿托品(10^-10和10^-9mol/L)能阻断同浓度ACh的增强作用;N型胆碱能受体阻断剂筒箭毒碱(10^-10和10^-9mol/L)可部分阻断同浓度ACh的增强作用。结论:在机体内副交感神经兴奋可导致T淋巴细胞的功能增强,其免疫调节作用由免疫细胞上的M和N受体介导,可能M受体的作用占主导地位。

Abstract:Objective:To investigate the effect of ACh on the Con A-induced IL-2 production . Methods: The single splenocytes from rats weresuspended in the complete culture medium and were stimulated with Con A. MTT assay was used to determine their IL-2 production. Results:The splenocytes treated with ACh at concentrations ranging from 10 -6 mol/L to10 -10 mol/L significantly enhanced Con A-induced IL-2 production compared with those induced by Con A alone. Muscarinic cholinergic receptor agonist pilocarpine and nicotinic cholinergic receptor agonist nicotine enhanced IL-2 production of rat splenocytes. The stimulating effect of ACh at 10 -10 mol/L concentration was completely blocked by muscarinic receptor antagonist atropin in 10 -9 mol/L;the effect induced by 10 -9 mol/L ACh was partially prevented by 10 -9 mol/L atropine. Tubocurarine, a nicotonic cholinergic antagonist, could also partially block IL-2 release induced by ACh. Conclusion:ACh in a wide range of concentrations enhanced the Con A-induced IL-2 production. The effect of ACh was mediated by M and N cholinergic receptors, and probably, M receptors played a major role in the regulation of IL-2 production.
