人参皂苷单体Rb1对豚鼠模拟缺血心室肌细胞动作电位的影响

摘    要:目的:观察人参皂苷单体Rb1对模拟缺血状态下豚鼠心室肌细胞动作电位的影响。方法:采用酶解法分离豚鼠心室肌细胞,利用全细胞膜片钳技术的电流钳模式记录正常和模拟缺血条件下心室肌细胞的动作电位的变化。结果:对于模拟缺组心肌细胞,Rb1能明显缩短动作电位时程并具有剂量依赖性(n=8,P〈0.05),而静息电位和动作电位幅值无明显改变。结论:Rb1对缺血心肌细胞的这种作用可能是其抗心肌缺血机制之一。

Abstract:Objective:Department of physiology.Methods:To investigate the effect of Rb1 on action potential in ischemic cardiomyocytes. Results:Single myocytes were dissociated by enzymatic dissociation method .The changes of action potential were recorded by current-clamp mode with whole patch clamp technique under normal and ischemic conditions. Results: Rb1 can significantly shorten action potential duration in ischemic cardiomyocytes in a concentration-dependent manner. (n=8,P<0.05).Resting potentials and amplitude of action potential were no changes.Conclusion:Rb1 may be one of mechanism on anti-myocardial ischemia.
