摘    要:目的:研究不同浓度葡萄糖对INS-1细胞及胰岛分泌功能的影响,并探讨氧化应激在葡萄糖对INS-1细胞功能损伤中的作用。方法:将不同浓度葡萄糖(11.1、16.7、22.2、33.3mmol/L)作用于INS-1细胞,镜下观察INS-1细胞的形态改变,应用可见分光光度计测定丙二醛(MDA)含量,运用ELISA方法检测葡萄糖刺激的胰岛素分泌(GSIS)。结果:随着葡萄糖浓度的增加,INS-1细胞的凋亡增多,MDA含量逐渐增加,GSIS值逐渐下降,不同浓度葡萄糖组间MDA含量、GSIS值差异有统计学意义(P均<0.01)。结论:高糖可促进INS-1细胞凋亡,在高糖作用下活性氧物质(ROS)代谢产物MDA增多,胰岛细胞胰岛素分泌功能受损。

Abstract:Objective: To determine the dose-dependent effect of glucose on the secretary activity of INS-1 cells,and to explore the role of oxidative stress in glucose-meidated functional damage of INS-1 cells.Methods: INS-1 cells were treated with glucose at concentrations of 11.1、16.7、22.2、33.3 mmol/L,then the morphological changes were observed under a light microscope.Malondialdehyde(MDA) content in the culture media was determined by spectrophotometry and the glusoe-stimulated insulin secretion(GSIS) was assessed by ELISA.Results:As the glucose dose increased,the INS-1 cell apoptosis increased,MDA concentration gradually elevated while the GSIS gradually decreased showing significant differences among groups treated with different concentration of glucose.Conclusions:Under high glucose conditions,the reactive oxygen species(ROS) metabolite MDA increases,leading to apoptosis and thereby impaired insulin secretion in isle
