牛蒡子苷对小鼠3T3-L1前脂肪细胞增殖与分化的影响及其作用机制

摘    要:【目的】探讨牛蒡子苷(arctiin)对小鼠3T3-L1前脂肪细胞增殖、分化的影响及其作用机制。【方法】采用不同浓度牛蒡子苷作用于3T3-L1前脂肪细胞,四甲基偶氮唑盐(MTT)法检测其对细胞增殖的影响;油红O染色及分光光度法分析脂肪细胞的分化程度;实时荧光定量PCR检测细胞过氧化物酶增殖物激活受体γ(peroxisome proliferators activatedreceptorγ,PPARγ)和CCAAT/增强子结合蛋白α(CCTTA enhancer binging proteinα,C/EBPα)的表达情况。【结果】牛蒡子苷对3T3-L1细胞增殖无显著性影响(P>0.05),但可显著抑制3T3-L1前脂肪细胞向成熟脂肪细胞分化(P<0.05或P<0.01),且呈一定剂量依赖关系;与空白对照组比较,牛蒡子苷可显著降低PPARγmRNA、C/EBPαmRNA表达水平(P<0.05或P<0.01)。【结论】牛蒡子苷能抑制3T3-L1前脂肪细胞的成脂分化,其机制可能与其能抑制脂肪细胞分化相关调控因子PPARγ和C/EBPα的表达有关。

Abstract:Objective To explore the effect and mechanism of arctiin on proliferation and differentiation of murine 3T3-L1 preadipocytes.Methods 3T3-L1 preadipocytes were cultured with arctiin in different dosages.Cell proliferation was analyzed by methyl thiazolyl tetrazolium colorimetric assay(MTT).The degree of cell differentiation was evaluated by oil red O staining and spectrophotography.The expression of peroxisome proliferators activated receptor γ(PPARγ) and CCTTA enhancer binding protein α(C/EBPα) was quantitatively detected by real-time fluorescence quantitative-RT-PCR.Results Arctiin showed no significant influence on the proliferation of 3T3-L1 preadipocytes(P>0.05),but inhibited 3T3-L1 preadipocytes differentiating into mature adipocytes in a dose-dependent manner(P<0.05 or P<0.01).The mRNA levels of PPARγ and C/EBPα were decreased by arctiin(P<0.05 or P<0.01 compared with the blank control group).Conclusion Arctiin can inhibit the differentiation of 3T3-L1 preadipocytes.The underlying mechanism may be related to inhibiting the expression of PPARγ and C/EBPα which are associated with cell differentiation.
