通肺胶囊对弹性蛋白酶诱发大鼠慢性阻塞性肺疾病模型的影响

【目的】观察通肺胶囊对弹性蛋白酶诱发大鼠慢性阻塞性肺疾病(COPD)模型的影响。【方法】选用SD大鼠,随机分为空白对照组,模型组,阳性对照组(盐酸班布特罗,剂量为3.3 mg.kg-1.d-1),通肺胶囊高、中、低剂量组(剂量分别为5.24、2.62、1.31 g.kg-1.d-1);除空白对照组外,其他组大鼠均以弹性蛋白酶诱发大鼠COPD模型,进行腹主动脉血的血气分析,测定动脉血血清及肺组织匀浆中丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性,进行右肺及支气管细胞学计数及分类检查,测定左肺体积及进行左肺组织病理学检查。【结果】通肺胶囊高、中、低剂量组均可显著提高模型大鼠血氧分压(P<0.05或P<0.01),通肺胶囊高剂量组可显著增加模型大鼠白细胞数量(P<0.01),通肺胶囊各剂量组均可显著降低模型大鼠血清MDA含量(P<0.05或P<0.01),通肺胶囊高剂量组可显著提高模型大鼠血清SOD活性(P<0.05),通肺胶囊中、低剂量组均可显著降低模型大鼠肺匀浆组织中MDA含量(P<0.05或P<0.01)。通肺胶囊各剂量组均可不同程度改善模型大鼠肺泡扩张、肺泡间隔断裂及肺泡融合等病理变化。【结论】通肺胶囊对弹性蛋白酶诱发的慢性阻塞性肺疾病模型大鼠具有一定的治疗作用。

Effect of Tongfei Capsule on Rat Model of Chronic Obstructive Pulmonary Disease Induced by Elastase

Objective To observe the effect of Tongfei Capsule on the rat model of chronic obstructive pulmonary disease(COPD)induced by elastase.Methods SD rats were randomized into blank control group,model group,positive control group(treated with Bambuterol Hydrochloride Tablet of 3.3 mg·kg-1·d-1),and high-,middle-and low-dose Tongfei Capsule groups(treated with Tongfei Capsule in the dosage of 5.24,2.62,1.31 g·kg-1·d-1 respectively).We established the rat model of COPD with elastase,and then performed blood gas analysis of abdominal aorta blood,determined serum malondialdehyde(MDA)content and superoxide dismutase(SOD)activity,and measured MDA content and SOD activity in the homogenate of lung tissue.In addition,we recorded the results of the cytological count and classification examination of right lung and bronchia,determined the volume of left lung and performed histopathological examination of left lung.Results In high-,middle-and low-dose Tongfei Capsule groups,blood oxygen pressure was increased and serum MDA content was decreased(P<0.05 or P<0.01).Serum SOD activity was enhanced(P<0.05)and the number of white blood cells was increased(P<0.01)in high-dose Tongfei Capsule group.In low-and middle-dose Tongfei Capsule groups,MDA content in lung tissue homogenate was decreased(P<0.05 or P<0.01).Alveolar ectasia,alveolar septum rupture and alveolar fusion were improved to various degrees in Tongfei Capsule groups.Conclusion Tongfei Capsule exerts certain therapeutic effect on the rat model of chronic obstructive pulmonary disease induced by elastase.
