一株海南红树林真菌胞外多糖抗病毒活性初步研究

摘    要:目的:对分离自海南红树林真菌菌株PH1018的胞外多糖体外抗流感病毒H3N2和柯萨奇B组3型病毒(CoxB3)活性进行研究。方法:收集纯化菌株PH1018产生的胞外多糖,观察其细胞毒性和对抗病毒引起的细胞病变效应。菌株分类采用形态学观察和ITS序列分析。结果:多糖样本可明显抑制流感病毒H3N2,细胞病变半数抑制浓度为34.32μg/mL,选择指数达到41.3。CoxB3的IC5 0为7.81μg/mL,选择指数达到212。菌株初步鉴定为杂色曲霉。结论:从海南红树林分离一株杂色曲霉,其产生的胞外多糖在体外具有抗流感病毒H3N2和CoxB3活性,值得进一步研究。

Abstract:Objective: To explore antiviral activities of exopolysaccharide(EPS)from one kind of fungi strain PH1008 isolated from mangrove in Hainan province against H3N2 influenza virus and coxsackievirus(CoxB3) in vitro.Methods:The EPS of strain PH1018 was collected and purified.The toxicity and antiviral actions of EPS was evaluated by observing drug cytotoxicity effect and cytopathic effect(CPE).The taxonomic status of strain was identified by morphological observations and analysis of the ITS sequence.Results:The EPS inhibited H3N2 influenza virus at the IC50 values of 34.32 μg/mL,and SI values of 41.3.The EPS inhibited CoxB3 at the IC50 values of 7.81 μg/mL,and SI values of 212.The strain was identified as Aspergillus versicolor.Conclusions:EPS from fungi strain PH1018 isolated from mangrove in Hainan province has remarkable anti H3N2 influenza virus and CoxB3 activities in vitro,this EPS could be utilized for the further research.
