白芍总苷的降血脂、抗氧化作用及其对脂肪肝的防治研究

摘    要:目的:研究白芍总苷的降血脂、抗氧化作用及其对脂肪肝的防治作用。方法:采用高脂高糖饮食建立脂肪肝大鼠模型,并预防性给予白芍总苷处理8周。8周后,处死大鼠,测定血浆中总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、丙氨酸转氨酶(ALT)、天门冬氨酸转氨酶(AST)水平;取全肝称重,计算肝指数,同时测定肝组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-PX)活性以及丙二醛(MDA)的含量,取肝左叶,HE染色,光镜下观察肝脏组织学变化。结果:白芍总苷可明显降低脂肪肝大鼠血浆TC、TG、LDL-C水平,提高血浆HDL-C水平;降低肝指数,降低ALT和AST水平,明显提高肝脏组织中SOD、CAT和GSH-PX活性,降低MDA含量,改善肝脏组织学改变。结论:白芍总苷对高脂高糖饮食诱导的脂肪肝有防治作用,其作用机制可能与降血脂和提高肝脏抗氧化能力有关。

Abstract:Objective: To study hypolipidemic and antioxidative effect and hepatic protective effect of total glucosides of paeony′s on non-alcoholic fatty liver.Methods:Rats were bred with high sugar and fat diet to establish non-alcoholic fatty liver model.Total glucosides of paeony(0.1 g/kg) was administered to prevent development of non-alcoholic fatty liver.After 8 weeks,rats were sacrificed and the blood was collected to test total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C) levels and the activities of aspartate aminotransferase(AST),alanine aminotransferase(ALT).Isolated liver was weighed to calculate liver index and used to evaluate the activities of glutathione peroxidase(GSH-PX),superoxide dismutase(SOD),catalase(CAT) and the amount of malonaldehyde(MDA) as well as histological changes.Results:Total glucosides of paeony can decrease TG,TC,LDL-C,increase HDL-C,lower the activities of ALT,AST and liver index of non-alcoholic fatty liver rats,in addition,total glucosides of paeony can enhance activities of SOD,CAT,GSH-PX,diminish amount of MDA greatly and improved histological changes in liver of non-alcoholic fatty liver rats.Conclusion:Total glucosides of paeony can decrease serum lipid and improve hepatic function of non-alcoholic fatty liver rats;furthermore,the anti-oxidative and hypolipidemic effect may be responsible for its liver protective effect.
