Expression level of GPC1 in HCC and its clinical prognostic significance

Objective:To investigate the expression level of GPC1 in patients with HCC and their cell lines,and its correlation with clinical prognosis.Methods:From October 2011 to November 2013,175 patients with HCC and 27 patients with hepatic hemangioma(HC)were diagnosed in our hospital.All patients were confirmed by pathological examination and underwent radical resection of liver cancer by laparotomy or laparoscopy.The follow-up period was up to December 2018,with a median follow-up of 43 months.The expression levels of GPC1 in HCC tissues and matched paracancerous tissues,serum and hepatoma cell lines were detected by RTQ-PCR,Western blot(WB),immunohistochemistry and ELISA.At the same time,the correlation between the expression level of GPC1 and the poor prognosis was analyzed.Results:The expression of GPC1 was up-regulated in HCC and stronger in metastatic HCC.The same result was also observed in HCC cell line.The serum GPC11 of HCC patients was higher than that of healthy control group(HC),the difference was statistically significant(P<0.05).The recurrence free survival rate(RFs)and disease-specific survival rate(DSS)of HCC patients in high GPC1 expression group were significantly lower than those in low GPC1 expression group(P<0.05).In addition,the high expression of GPC1 was significantly correlated with tumor size and TNM stage(P<0.05).Tumor size,TNM stage and GPC1 expression were independent predictors of RFS and DSS in HCC patients.Conclusion:The high expression of GPC1 in HCC patients is closely related to the poor prognosis.GPC1 can be used as a potential diagnosis and treatment index in HCC patients。
