漳州市肠道病毒71型分离株的VP1区基因特征分析

摘    要:目的了解漳州市肠道病毒71型分离株的VP1区基因特征。方法对2010年漳州市的5株肠道病毒71型分离株进行VP1区基因全长序列测定,测序结果利用DNASTAR软件进行核苷酸、氨基酸序列分析和同源性比较,并用Mega4.0软件构建系统发生树。结果5株肠道病毒71型分离株的VP1区核苷酸序列全长均为891bp,且与2008年安徽阜阳流行株Fuyang.Anhui.P.R.C-17.08-2的VP1区核苷酸序列同源性和氨基酸序列同源性最高,分别为98.2%-98.8%和99.7%-100.0%。5株肠道病毒71型分离株间的核苷酸序列同源性为97.2%-99.8%.氨基酸序列同源性为99.3%~100.0%。氨基酸在98位和218位这两个位点发生了特异性变异。VP1区基因遗传进化分析表明,漳州市所有分离株均属于C4基因亚型的C4a进化分支。结论5株肠道病毒71型分离株均属于C4基因亚型的C4a进化分支.与2004年以来的中国大陆EV71病毒流行的基因型完全-致。

Genetic characterization of the VP1 region of enterovirus 71 isolated in Zhangzhou city

Abstract:Objective To analyze the genetic characteristics of the VP1 region of enterovirus 71 (EV71 ) isolated in Zhangzhou city. Methods EV71 were isolated from the specimens of hand-foot-and-mouth disease (HFMD) patients in Zhangzhou in 2010. Complete VP1 genes of the five EV71 isolates were sequenced. DNAStar was used to analyze the degree of nucleotide sequence and amino acid sequence homology, and Mega 4.0 was used to construct the phylogenetic tree. Results Sequence analysis revealed that the full-length VP1 of all the five EV71 isolates had the same number of base pairs (891 bp). The degree of nucleotide and amino acid sequence homology between the isolates and the Fuyang.Anhui.P.R.C-17.08-2 strain was 98.2%-98.8% and 99.7%-100.0%, respectively. The degree of nucleotide and amino acid sequence homology among the five isolated strains was 97.2%-99.8% and 99.3%-100.0%, respectively. Mutations were found at the amino acid position 98 and 218. Full-length VP1 and genome-based phylogenetic analysis revealed that the Zhangzhou strains belonged to the same subgenotype of C4a. Conclusion Five EV71 isolated in Zhangzhou city were phylogenetically related to C4a in the C4 subgenotype. The genotype of these isolates is in line with the EV71 circulating in mainland China since 2004.
