炮制对王不留行中刺桐碱及黄酮苷类成分含量及溶出率的影响

摘    要:目的:研究炮制对王不留行中刺桐碱及黄酮苷类成分含量的影响,探索炮制对王不留行主要化学成分及其溶出率的影响。方法:采用HPLC测定炮制前后王不留行中刺桐碱、王不留行黄酮苷、肥皂草苷、异牡荆素-2″-O-阿拉伯糖苷和王不留行黄酮苷H的含量及水煎溶出率,流动相乙腈(A)-水(B)梯度洗脱(0~12 min,13%A;12~18 min,17%A;18~30 min,20%A),检测波长280 nm。结果:炮制前,这5个成分的质量分数分别为0.13%,0.70%,0.018%,0.051%,0.11%,炮制后则分别为0.12%,0.52%,0.016%,0.098%,0.091%;5个成分的水煎溶出率分别升高了7.7%,10.2%,10.8%,11.4%,17.8%。结论:炮制后异牡荆素-2″-O-阿拉伯糖苷含量有较大幅度上升,而王不留行黄酮苷含量有较大下降,其余成分含量有少量下降。炮制对王不留行中刺桐碱及黄酮苷类成分含量的影响存在一定差异,但炮制后这些成分的水煎溶出率均上升。

Abstract:Objective: To investigate the influence of processing on contents of hypaphorine and flavonoid glycosides in Vaccariae Semen. Method: Before and after processing,HPLC was employed to determined contents and dissolution rates of hypaphorine,vaccarin,saponarin,isovitexin-2"-O-arabinoside and vaccarin H in Vaccariae Semen with the mobile phase of acetonitrile-water for gradient elution and detection wavelength at 280 nm. Result: Before processing,their contents were 0.13%,0.70%,0.018%,0.051%and 0.11%;after processing,their contents were 0.12%,0.52%,0.016%,0.098%and 0.091%,respectively.However,dissolution rates of these five ingredients were raised by 7.7%,10.2%,10.8%,11.4%and 17.8%,respectively. Conclusion: Processing has different influence on contents of hypaphorine and flavonoid glycosides in Vaccariae Semen,but their dissolution rates raise.
