摘    要:目的比较piggyBac(PB)转座子和Tol2转座子介导基因整合到细胞基因组中并实现长期表达的能力,探索将转座子作为一种研究基因长期表达的工具。方法采用荧光蛋白和分泌型碱性磷酸酶(secretedalkalinephosphatase,SEAP)作为报告基因,利用PB转座子和Tol2转座子在CHO细胞和HEK293T细胞中进行细胞转染实验,在传代前和传代后的不同时间点持续监测报告基因表达量的变化情况。结果与不加转座酶的对照组相比,瞬时转染CHO细胞和HEK293T细胞后,PB转座子和Tol2转座子能促进携带的荧光蛋白和SEAP报告基因的表达,并且能够长期表达,PB转座子介导报告基因长期表达的效果优于T012转座子。结论PB转座子介导基因长期表达的能力强于Tol2转座子,PB转座子更适于研究基因的长期表达。

Abstract:Objective To develop transposon as a tool for researches on long-term gene expression by comparing the capabilities of long-term gene expression mediated by piggyBac ( PB ) transposon and Tol2 transposon. Methods PB transposon and Tol2 transposon, which express the reporter gene of fluo- rescent protein or secreted alkaline phosphatase (SEAP) , were transfected into CHO cells and HEK 293T cells. Then , the expressions of fluorescent protein and SEAP were detected at a series of time points both before and after cells passaged. Results After PB and Tol2 transposons were transiently transfected into CHO cells and HEK 293T cells, the expression cells that transfected with the plasmids including levels of fluorescent protein and SEAP were higher in the transposase than those without transposase. Though both of them had expressed the reporter genes for a long time, PB transposon was more efficient than Tol2 transposon. Conclusion Our study suggests that PB transposon is of more efficiency than Tol2 transposon in mediating long-term gene expression.
