摘    要:目的探讨骨髓间充质干细胞(BMSCs)对血管性痴呆(VaD)大鼠的作用。方法以两血管法制备VaD模型。将大鼠随机分为假手术组、磷酸盐缓冲液组、BMSCs组。造模1周后,尾静脉注射BMSCs;4周后行Morris水迷宫检测,观察其行为学改变;免疫组化法检测大鼠海马区神经细胞NGF、SYN表达。结果与PBS组比较,BMSCs组逃避潜伏期明显缩短,SYN与NGF表达明显升高(P0.05或P0.01)。结论 BMSCs对VaD大鼠的学习记忆能力有一定改善作用,可提高NGF及SYN表达。

Abstract:Objective To investigate the effect of bone marrow mesenchymal stem ceils (BMMSCs) on rats with vascular dementia(VaD). Methods VaD models were randomly divided into 3 groups: sham-operation group, phosphate buffer solution group (PBS group)and BMSCs group. 4 weeks later, behavioral abnormalities were observed by using Morris water maze test and the expression of NGF and synaptophysin (SYN) in hippocampus neurons was detected by immunohistochemistry. Results Morris water maze test showed that the escape latency in the BMSC group was significantly shorter than that in the PBS group, and the expression of NGF and SYN was significantly higher than that in the PBS group (P〈0.05 or P〈0.01 ). Conclusion BMSCs can improve abilities of VaD rats to learn and memory and increase NGF and SYN expression in rats with VaD.
