羟基积雪草苷大鼠体内代谢研究

摘    要:目的:研究羟基积雪草苷在大鼠体内的主要代谢产物,寻找羟基积雪草苷体内药效物质。方法:采用高效液相色谱一电喷雾串联质谱(HPLC—ESI—MS“)法鉴定大鼠腹腔注射羟基积雪草苷(50mg·kg-1)后体内的代谢产物。结果:从大鼠含药血浆、尿液及脑组织中检测到羟基积雪草苷原形成分和11种代谢产物,均为相代谢产物。结论:羟基积雪草苷在大鼠体内代谢广泛,主要的代谢途径为去甲基化、羟基化、脱水等反应。

Abstract:Objective: To identify madeeassoside (MC)and its metabolites in rats. Methods: A high- performance liquid chromatography-electrospray ionization tandem mass spectrometry (HPLC-ESI-MSn) method was applied to analyze the metabolites in plasma, urine and brain after a single intraperitoneal administration of 50 mg-kg-1 MC to rats. Results : MC and eleven I phase metabolites were detected in plasma, urine and brain. Conclusion : MC undergoes extensively metabolism in rats. The major metabolic pathways were demethylation, hydroxylation dehydration.
