乳香中三萜类成分的薄层色谱鉴别和含量测定

摘    要:目的:以乳香中主要三萜类成分为指标,建立和完善乳香的定性定量分析方法。方法:用TLC鉴别法,以三萜酸类成分11-羰基-β-乙酰-乳香酸(AKBA)为对照品,采用硅胶GF254薄层板,以环己烷-乙酸乙酯-冰醋酸(10:2:0.2)为展开剂,鉴别乳香药材中AKBA;用HPLC测定方法,Agilent TC-C18色谱柱(250mm×4.6mm,5μm),乙腈-0.05%磷酸溶液(80:20)为流动相,流速:0.8mL·min^-1,检测波长:250nm,测定不同来源乳香药材中AKBA和11-羰基-β-乳香酸(KBA)含量。结果:乳香TLC图谱中,AKBA斑点清晰、分离效果好;HPLC含量测定法中,AKBA和KBA分别在0.02463~0.2463mg·mL^-1和0.01004~0.1004mg·mL^-1,线性关系良好。AKBA和KBA的回收率分别为96.1%(RSD=3.1%)和96.4%(RSD=3.5%)。结论:所建立的TLC和HPLC法操作简便、结果准确、重复性好,可作为乳香定性、定量的质量控制方法。

Abstract:Objective:To establish and improve the qualitative and quantitative analysis of triterpenes in the Olibanum. Methods: TLC was used for qualitative analysis of triterpenes in the Olibanum, silica gel GF254, mixed with sodium carboxymethyl cellulose as the coating substance and cyclohexane-ethyl acetate-glacial acetic acid (10:2:0. 2)as mobile phase,and triterpene acids composition of Olibanum as reference substance namely acetyl-11-keto-β-boswellic acid (AKBA). HPLC method was used for quantitative analysis of AKBA and 11 -keto-β-boswellic acid ( KBA), using Agilent TC-C 18 column(250 mm ×4. 6 mm ,5 μm) . The mobile phase was a mixture of acetonitrile-0.05 % phosphoric acid(80:20) . The flow rate was 0. 8 mL-min-1. The column temperature was 30 ℃. The detection wavelength was 250 nm. Results:The TLC method had high sensitivity and showed clear spots of AKBA. The recoveries of AKBA and KBA by HPLC method were 96. 1% ( RSD = 3.1% ) and 96.4% ( RSD = 3.5% ), respectively. Conclusion: TLC and HPLC methods are simple, accurate and repeatable. The methods can be used for the quality control of Olibanum.
