高脂饮食诱发胰岛素抵抗大鼠模型的建立与评价

摘    要:目的建立饮食性胰岛素抵抗大鼠模型及评价方法。方法高脂饲料喂养不同年龄和体质量Wistar大鼠,4周后检测血糖、血脂、游离脂肪酸和胰岛素水平并行高胰岛素-正葡萄糖钳夹实验。结果青年高脂组较青年对照组、老年对照组较老年高脂组相比,高脂喂养大鼠体质量虽有所增加,但差异无统计学意义(P均〉0.05)。青年高脂组与青年对照组、老年高脂组与老年对照组、老年对照组与青年对照组及老年高脂组与青年高脂组相比,血清总胆固醇、血清甘油三酯及空腹胰岛素升高,空腹血糖升高,葡萄糖输注率下降,有显著性差异(P〈0.05或0.01)。结论高脂饲料喂养青年和老年大鼠,均可成功建立胰岛素抵抗大鼠模型,钳夹实验用于胰岛素抵抗大鼠模型的评价更准确、可靠。年龄和体质量因素与胰岛素抵抗有关。

Preparation and evaluation of insulin resistance model in rats with high - fat diet

Abstract:Objective It is to establish and evaluate the insulin resistance model in rats with high-fat diet. Methods Male Wistar rats aged 4 -5 months were divided into young control (YC) group (n = 10) and young high -fat diet .(YHF) group ( n = 10 ) , Male Wistar rats aged 22 - 24 months were divided into old control ( OC ) group ( n = 10 ) and old high-fat diet (OHF) group ( n = 10). Four weeks later, Body Weight was observed and the serum was separated for the detection of lipid, Glucose, FFA, insulin. The hyperinsulinaemie-euglycaemic clamp was performed to confirm that the insulin resistant rat model was duplicated successfully. Results After four - week high fat diet, Body weight was higher in YHF group than that in YC group, but was not different between the two groups (P 〉 0.05). That was higher in OHF group than that in OC group, but was not different between two groups too ( P 〉 0.05 ). Compared with YC group serum total cholesterol, triacylglycerol, fasting insulins and FFA increased in YHF group (P 〈 0.05 or 0.01 ) ; Compared with OC group serum total cholesterol, triacylglycerol, fasting insulins and FFA increased in OHF group (P 〈0.05 or 0.01 ) ; Compared with YC group serum total cholesterol, triacylglyeerol, fasting insulins and FFA increased in OC group (P 〈 0.05 or 0. 01 ) ; Compared with YHF group serum total cholesterol, triaeylglyeerol, fasting insulins and FFA increased in OHF group (P 〈 0.05 or 0. 01 ). compared with the YC group fasting blood glucose increased and glucose infusion rate decreased in YHF group (P 〈0.05 or 0.01 ) ; Compared with OC group fasting blood glucose increased and glucose infusion rate decreased in OHF group (P 〈 0.05 or 0.01 ) ; Compared with YC group fasting blood glucose increased and glucose infusion rate decreased in OC group (P 〈 0.05 ) ; Compared with YHF group fasting blood glucose increased and glucose infusion rate decreased in OHF group (P 〈 0.05 or 0.01 ). Conclusion The insulin resistant model can be duplicated successfully in young and old rats with high-fat diet; the hyperinsulinaemic-euglycaemic clamp technique is accurate and reliable in evaluating insulin resistance in rats. Age and weight are related to the induction of insulin resistance.
