正常妊娠及妊高征患者外周血中IL-12、IL-18的检测意义

目的:检测非孕妇女、正常妊娠及妊娠高血压综合征(妊高征,PIH)患者外周血中IL-12和IL-18水平,探讨Th1/Th2细胞因子在妊高征发病中的作用。方法:孕20周后的子痫前期患者34例(轻度子痫前期15例,重度子痫前期19例),正常妊娠组32例,非妊娠对照组16例。取外周血进行流式细胞仪检测,选取CD28表达量最高者的淋巴细胞作为实验对象。采用ELISA法测定IL-12、IL-18,流式细胞仪测定Th1/Th2细胞比。结果:正常妊娠组、非孕组及妊高征组IL-12比较均存在显著性差异(P<0.001)。轻度子痫前期组、重度子痫前期组与正常妊娠组IL-18比较均不存在显著性差异(P>0.05)。各组IL-18/IL-12浓度比比较,正常妊娠组与妊高征组比较存在显著性差异(P<0.0001)。IL-18/IL-12浓度比与Th1/Th2细胞比的相关性比较,呈明显负相关(P<0.01)。结论:正常妊娠的IL-18/IL-12浓度比明显高于妊高征组与Th1/Th2细胞比呈相反变化。

The detection significance of interleukin (IL)-18 and IL-12 in peripheral blood of normal pregnant and pregnancy-induced hypertension patients

Objective:To investigate the levels of IL-18 and IL-12 in peripheral blood of normal pregnant and in pregnancy-induced hypertension patients,and to explore the role of Th1/Th2 cytokines in the pathogenesis of pre-eclampsia.Methods:34 cases pre-eclampsia patients pregnant 20 weeks(mild pre-eclampsia 15 cases and severe pre-eclampsia 19 cases),normal pregnancy group 32 cases,non-pregnant controls 16 cases.The peripheral blood was tested by flow cytometry,selected the lymphocytes with highest expression of CD28 as experimental subject.IL-12 and IL-18 were detected by ELISA Th1/Th2 ratios in PBMC was detected by flow cytonetry.Results:IL-12 in normal pregnancy group,pregnancy-induced hypertension group and non-pregnant group were significant difference(P<0.001).IL-18 in mild pre-eclampsia cases,severely pre-eclampsia cases and normal pregnancy subjects had no significant difference(P>0.05).While the ratios of IL-18/IL-12 in normal pregnancy group and pregnancy-induced hypertension group had significant difference(P<0.000 1).Thl/Th2 ratios were negatively correlated with the ratios of IL-18/IL-12.Conclusion:The ratios of IL-18/IL-12 were significantly higher in normal pregnant group than pregnancy-induced hypertension group,and Th1/Th2 ratios were negatively correlated with the ratios of IL-18/IL-12.
