高血压脑出血患者血肿周围水肿组织中水通道蛋白-4表达及意义

摘    要:目的研究高血压脑出血患者血肿周围水肿组织中水通道蛋白-4(aquaporin 4,AQP4)表达及其意义。方法收集2009~2010年高血压脑出血患者血肿周围水肿组织28例,按照脑出血时间分为四组,如<12h,≥12h且<24h,≥24h且<48h和≥48h且<72h,以脑外伤患者术中急性脑膨出行内减压而切除的正常脑组织作为对照,应用Western-Blot方法,以GAPDH为内参,分别检测血肿周围水肿组织及正常脑组织中AQP4的表达水平。结果脑出<12h,≥12h且<24h,≥24h且<48h和≥48h且<72h患者脑内血肿周围水肿组织AQP-4的表达水平明显高于对照组(P<0.05),72h内AQP-4的表达逐渐升高。结论高血压脑出血患者脑内血肿周围水肿组织AQP4的表达增多与脑水肿的形成和发展密切相关。

Expression and significance of aquaporin-4 in edematous brain tissue around hematomas after hypertensive intracerebral hemorrhage

Abstract:Objective To study the early aquaporin-4 (AQP4) expression in edematous brain tissue around hematomas after hypertensive intraeerebral hemorrhage. Methods The expressions of AQP4 in the edematous brain tissues around hematomas in 28 patients with hypertensive intracerebral hemorrhage from 2009-2010 were studied. The patients were divided into four groups according to the time after the hemorrhage, i.e. 〈 12 h group(A group),≥12 h and 〈24 h group ( B group),≥ 24 h and 〈 48 h group ( C group), ≥ 48 h and 〈 72 h group. Normal brain tissue was from acute encephalocele patients who need inline decompression and resection in operations of traumatic brain injury patients. Levels of AQP4 expression in edematous brain tissues around the hematoma and normal brain tissue were detected separately by using Western-Blot with GAPDH as an internal. Results The expression levels of AQP4 in the edematous cerebral tissues around the hematomas at A, B, C, D groups after the hemorrhage were significantly higher than that in the control group ( P 〈 0. 05 ). And the AQP-4 expression was gradually increased within 72 h. Conclusion The increased tissue expression of AQP4 in hypertensive cerebral hemorrhage edema of intracerebral hematoma is closely related to brain edema formation and development.
