噬菌体展示抗体库技术的研究进展及临床应用

摘    要:治疗性抗体的发展,主要经历了异源性抗体、人源化抗体和人源性抗体几个阶段。目前,人源性抗体是治疗性抗体发展的主要方向,而噬菌体抗体库构建技术为人源性抗体的制备提供了良好的技术平台。噬菌体展示抗体库构建技术,是一种将抗体组合文库与噬菌体表面展示技术相结合所形成的新技术,是20世纪90年代初期抗体工程领域的一项重大进展,目前广泛应用于生物医学的多个方面。将就此技术的原理、分类、筛选方法及主要应用领域等方面进行综述。

Progress and its clinical application of phage display antibody library technology

Abstract:The development of therapeutic antibodies experienced three stages, including stage of heterologous antibodies, humanized antibodies and fully human antibodies. At present, the research of fully human therapeutic antibodies is the key direction of its development, and phage antibody library technology provides a good technological platform for preparation of fully human antibodies. Phage display antibody library is the combination of antibody library and phage display technology, and is a significant advance in the field of antibody engineering in the early 1990s and has been widely used in many fields of biomedicine. In this paper, the principle, classification, screening methods and main application fields of this technology are reviewed.
