高分辨率熔解曲线分析技术及其应用进展

摘    要:高分辨率熔解曲线(high-resolution melting,HRM)是一种基于单核苷酸熔解温度不同而形成不同形态熔解曲线的基因分析新技术,具有极高的敏感性,可以检测出单个碱基的差异,并且成本低、通量高、速度快、结果准确、不受检测位点的局限,实现了真正的闭管操作。在突变扫描、单核苷酸多态性分析、甲基化研究、基因分型、序列匹配等方面HRM分析技术发挥着重要作用。

Abstract:High-resolution melting curve(HRM) is a new genetic analysis technology based on single nucleotide polymorphism in the formation of different forms of melting curves.It has a very high sensitivity,which can detect single nucleotide differences.Meanwhile,it has a great deal of other advantages such as low cost,high throughput,rapid,accuracy and not subject to the limitations of testing sites which realizes closed tube operation.HRM analysis technology is playing an important role in the fields of mutation scanning,single nucleotidepolymorphism analysis,methylation studies,genotyping,sequence matching and other aspects.The development of this method will surely have a far-reaching impact on the study of human genes.
