江苏地区汉族人群八个短串联重复序列基因座遗传多态性研究

摘    要:目的:了解江苏地区汉族人群D14S306、D1S818、D3S1338、LPL、F13B、F13A01、FESFPS和vWA等八个短串联重复序列(STR)基因座的遗传多态性,并获得该群体的遗传学数据。方法:采用Chelex—100法从江苏地区无亲缘关系的汉族个体血标本122份中提取DNA。特异引物聚合酶链反应。产物经6%变性聚丙烯酰胺凝胶电泳分离,银染检测。结果:这八个STR基因座在江苏地区汉族人群均具有遗传多态性,并符合Hardy—Weinberg定律,它们的个人识别率范围从F13B的0.6869到vWA的0.9285,非父排除率范围从F13B的0.2499到vWA的0.6026不等。结论:为应用这八个STR遗传标记在中国江苏地区汉族群体中进行亲子鉴定和个人识别提供了概率计算依据和群体遗传学数据。

Abstract:Objective:To understand the genetic polymorphism at STR loci D14S306, D1S818, D3S1338, LPL, F13B, F13A01, FESFPS and vWA for the Han population in Jiangsu province of China and construct a preliminary database. Methods: Allele and genotype frequencies for six terameric short tandem repeat (STR) loci were established from 122 unrelated Chinese Han individuals in Jiangsu province of China using locus-specific prime polymerase chain reactions (PCR), followed by electrophoresis of the PCR products in denaturing polyacrylamide gels and subsequent detection by silver staining. Results: All loci exhibit a high degree of polymorphism in the Han population in Jiangsu province of China and met Hardy-Weinberg expectations. The discrimination power (DP) ranged from 0.686 9 for F13B to 0.928 5 for vWA and the power of exclusion (PE) ranged from 0.249 9 for F13B to 0.602 6 for vWA. Conclusion: The obtain date can be highly suitable for forensic individualization and paternity tests to Han population in Jiangsu province of China.
