硒化壳聚糖诱导急性早幼粒白血病细胞凋亡的研究

摘    要:目的 研究硒化壳聚糖对体外培养的NB4细胞增殖和凋亡的影响,探讨这种作用与端粒酶活性和survivin蛋白的关系。方法MTT法检测细胞增殖;细胞形态学观察和流式细胞仪检测细胞凋亡;PCR-ELISA法检测端粒酶活性;western blot法检测survivin蛋白含量。结果50~200mg/L硒化壳聚糖可时间剂量依赖性地抑制NB4细胞增殖,诱导其凋亡,降低端粒酶活性和下调survivin蛋白含量。结论 硒化壳聚糖可通过抑制端粒酶活性,下调survivin蛋白含量,诱导细胞凋亡从而抑制体外培养的NB4细胞增殖。

Abstract:Objective To investigate the effect of selenium chitosan on proliferation and apoptosis of NB4 cells in vitro and to explore its relationship with the telomerase activity and survivin protein abundance. Methods: MTT was used to determine the inhibiting proliferation effect; the inducing apoptosis effect was observed by using microscope and flow cytometry; Telomerase activity was measured by polymerase chain reaction enzyme-linked immunoassay (PCR-ELISA); western blot was used to examine the abundance of survivin protein. Results: Exposure of NB4 cells to 50~200mg/Lof selenium chitosan for 24~72h produced a dose and time dependent increase in inhibiting rate and apoptosis rate as well; The telomerase activity and the abundance of survivin protein was strongly down regulated in selenium chitosan treated NB4 cells. Conclusions: selenium chitosan could inhibite NB4 cells proliferation in vitro by inducing cells apoptosis. decreasing telomerase activity and dowing regulation the abundance of survin protein.
