| 摘 要： | 目的:建立广泛表达红色荧光蛋白(RFP)的转基因小鼠,便于组织和细胞移植研究.方法:把DsRed基因片段插入到pCAGGS载体的强启动子下游构建转基因载体pCAG-DsRed.经细胞转染验证,受精卵原核注射,建立红色荧光蛋白转基因C57BL/6小鼠.PCR鉴定转基因小鼠的基因型;红色荧光蛋白在组织中的表达情况,则通过荧光显微镜检测小鼠各组织器官冰冻切片.结果:pCAG-DsRed载体可以在真核细胞中表达RFP,经基因型分析和表型观察,得到两只红色荧光蛋白的首建鼠.经荧光显微镜观察各组织冰冻切片发现红色荧光蛋白在多个组织器官中表达,其中在神经系统和肌肉组织中表达量较高.结论:成功建立了一种红色荧光蛋白转基因小鼠,红色荧光蛋白在转基因小鼠的神经系统和肌肉组织等中高表达.

| Abstract: | AIM:To establish a ubiquitously expressed red fluorescent protein (RFP) transgenic mouse model for tissue and/or cell transplantation. METHODS:The transgenic vector pCAG-DsRed was constructed by inserting the DsRed fragment into the downstream of the strong promoter in the pCAGGS vector and was validated by cell transfection.
